Bod, I assume that you just lightly slide the loop across the surface when you streak? Not putting any real pressure, not leaving a dent or a cut.....

I've seen your video using the loop with the print, but are you using a syringe for the initial drop directly onto plate then Z smear with loop? Or where are you getting solution from?

Squirting onto the loop. Or squirt spore solution into a sterilized cup like a shot glass then dip in that

Thanks Bod for the DIY inoculation loop. It seems a better way to get the spores up from the foil. Any spores left will use the 1mm shringe with droplets of distilled water and experiment with remaning spores
Heat the loop and cool in distilled water? is that okay to use
Also, have 20awg audio tinned wire? could this be used? it relatively sturdy, otherwise i can get approxiate wire here in Australia
Few cheep loops on ebay, but shipping ups the price

Bout time I upgraded my loop. Currently using 2 paper clips bent into a loop and wound on a pencil, taped, and PC'd. Hella ghetto but it works

Alright everyone. I know Bod suggests using whatchu got for simplicity, but I did some research, and it turns out 25 or 26 g kanthal or nichrome 80 would be optimal. The thinness guarantees better cooldown times. I noticed my current one made with 20g kanthal sizzles my agar during cooldown for nearly 30 sec. As we all know, exposure time matters in this hobby.
I also posted a link earlier about another tool, a Fungus Hoe, that seems to be perfect for ripping and transferring mycelium to new medium. Here's an updated link. What do y'all think?

I used stainless steel picture hanging wire from the hardware store for my first loop. Works great, and I still use it.
The pics in your that link you posted are broken BTW.

Since I've been dragging my feet on making an introduction thread, maybe my first post can be a contribution.
One way to scavenge Nichrome wire is from wirewound electrical resistors. They are typically found as the rectangular ceramic type meant to handle a lot of current and heat. I broke open a 10 ohm/10 watt power resistor and it had about 14" of 32awg Nichrome. Lower ohms with higher wattage ratings would probably have thicker wire.
I wouldn't suggest buying resistors just for the wire, but if you already have some...

Great stuff Bod thank you. I spent an hour earlier inoculating with one of those little "1 microliter" loops you mentioned, and afterwards started to worry that it might have been too small for the job. I bought them in complete noob ignorance from Ebay.
Three quick questions please:
1. Being plastic and not able to be flamed, do I need to use a different loop with each petri?
2. Can I PC the used loops to re-sterilise them?
3. Is there any advantage to using the pre-sterilised loops over following your DIY loop tek?
