BOD's DIY Inoculation Loop TEK 13

This tek is part of bod's cultivation is easy as fuck link listBod's DIY Inoculation Loop TEK Using X-Acto style handles and nichrome wire or easily found as vape coil wire You Will Need Some wire, experiment with different lengths 3.5-4 inches works well for me. some sort of thin rod to make the loop with, I used a thermometer and a handle, I used the x-acto for the clamping mechanism. This is how we doTake the x-acto knife apart fold a wire over to make a U shape and put the pointy ends into the blade receiver of the x-acto handle   now you have one of these    put your thin rod thing through, then push the wire so that it forms a loop around the utensil before you start to twist   if you don't do that  ugly mess if you spend the time to smooth it out    I also buy sterile disposable ones with much smaller loops 1 microliter(uL) they will both work for streaking spores out. With these ones you can bend your loop's eye to make it smaller to hold onto a drop of solution. for example take this  then crimp it into  this     Flame away

I do this too! I use a bread tie, remove the paper /plastic edges. Awesome once again sir!

Can contams get in the twists in the loop though? Do you always flame the entire thing?

in my agar tek I talk about how I keep a jar of 91% iso like a baby food jar. I dip my tools in there first and then flame them red hot. this way flame gets into all the crevices on tools. This is how a glass spreading rod would be sterilized since you can't hold glass in a flame till it's red hot and then touch a petri dish, it won't cool down fast enough. And it will eventually shatter too.

Nope 91 is more flammable burns hotter. And i would imagine has lower surface tension to get in more places

butane burns at 3578°F. 91% burns at 750°F.
70% is proven the more effective sterilizer. and the cracks and crevices get red hot from your butane torch.
so I guess im just not seeing the reasoning here. are you not using butane? if not, why not?

Propane torch anyway. 91% is just always what I've seen used for flaming glass rods. You're not using the alcohol to sanitize

Really simple, but great. I have an exacto sitting around since I switched to an actual scalpel. Will be trying this even though regular wire worked just fine. This looks a bit more pro/easier to grab with gloves on.

I am very inexperienced here, and maybe this is a stupid question, but what is the reason you wouldnt just use the syringe to squirt a drop on the petri dish. Is it just to get the spores to more areas on the dish, or am i missing the point entirely?

That is the reason. You don't want spores all sloppy over a dish And the more spore solution you use the higher your chance of contamination. And too much spore solution makes all the growth happen irregularly all over the dish so it's harder to grab glean mycelium.

Nice write up!
This is exactly the same as the loops they sell on lots of cultivation sites (I think these come from China). Its just a handle and it comes with a bunch of nichrome wires
You can save yourself some $ doing it yourself

Cool tek Bod  This is the exact same thing I did, it works perfectly I didn't use braided wire though, it's there a reason for choosing the braided?
