For transfers you can let them grow out enough for you to take a wedge, sometimes more if the growth looks questionable and letting it grow out a little more can help you see what is going on more clearly. But take into account that letting a plate grow more can help you identify more easily desirable growth patterns, so it all really depends on what you're after or what you need at any given moment.
If you're after strain isolation, IMO it would be better to let the myc colonize at least 60% of the plate, so it's easier for you to distinguish sectors and growth patterns.
For LI, tiger drop, wedge to grain, etc... you'd want to let the plates colonize fully or nearly.

Checking back in here. I just finished the Tiger Drop onto sterilized rye from the one agar plate I made that I cut what I thought were contaminants out of it. I know I did it wrong but it seems to me I got lucky.

I noticed some blue bruising on the other side of the agar but I couldn't get a good picture through the glass.
This was from the agar plates that had chunks of meat in them, lol.

So lacking in the patience department, ain't ya.
Were the contams you cut out sporulating (green) mold by any chance? If that's the case you can be sure you also tiger dropped millions of mold spores into that jar.
Agar is not used to cut out contams, it is use to transfer clean wedges to new plates, away from contams.
What you did is half-assing shit

I don't think that it was green mold I think what I cut out was probably some herbs or something from the potato mix. Someone told me on this thread about cutting out a clean wedge and moving it to a new agar plate but I had already done it
I made a lot of mistakes along the way but I'm definitely learning.
I know it's half assed but it was more from a lack of time than of patience. Also the plate was just sitting in my drawer with healthy looking mycelium so I just figured why not give it a shot. If everything turns into a moldy mess at least I got some practice for the next round.

1tsp agar-agar 2 tsp potato flakes 1 drop out honey .5 cup h2o 2 drops green food coloring
It seems really thick. 3 days After inoc I'm worried the spores can't penetrate the agar.. Is this the right thinking?

Its not a matter of if they can penetrate. mycelium can grow through solid wood. Its more of if there is enough moisture present to aid in germination.

and its too thick? it should be thin as f.
or do you mean
1 tablespoon agar 2 tablespoons potato... which would make it way to thick but from the potato not agar.

If you want to weigh it out, for this amount of agar you will want about 2.5 grams. If your using powdered agar and don't have a scale, a level tsp (5ml) is roughly 2.5 grams. Remember that no matter how much you want to mix up, you want a ratio of 20 grams of agar to 1 liter of water.
I used agar powder..

Wood isn't really "solid"- mycelium eats through the bonding agents in the "grains" of the wood first then consumes the more "solid" areas. And on Agar it only colonizes the top as it won't run down through it.
