clarification on long term storage methods

I put my cultures to slants for storage and i know about every 6 months to a year you need to grow the culture out on agar and make another slant. is there a way to store a culture and have it be good for years and years? i read somewhere about like cryogenic freezing. is that really viable or practical?

using wood in the slant helps but I still think it's best to transfer every once in a while to keep the culture healthy.

I think about once a year would be enough.

I may not be the most knowledgeable, but I believe RR said that a proper slant, with some form of wood could last upwards of two years if kept in the refrigerator. one of the posts I've read quoting him said some slants can last up to 5 years in the refrigerator and one guy on here said that his lasted from 2009 to 2015. They shouldn't die out after 6 months.

right on, i think just because i use a lot i'd probably run through a slant with in a year but it'd be cool on my last transfer to just transfer it to another slant

What do you mean by this? When you say you'd probably run through a slant in a year?

Remember, slants are for long term storage (1+ years) , not for expanding cultures, so there is really no reason you should ever use up a slant, since all expansion would be done on dishes

so i thought the order of things was you have a slant that makes master cultures, then those master cultures are used to make transfers to petris that go to grain. once the master gets used up don't you go back to the original slant to make another master or do you just use the last sliver of the master to make another master? sorry for all the masters lmao

From the knowledge I have, you make your slant, when you need to grow you take a tiny amount from the slant to an agar tray, put the slant back into the fridge. grow that tray  out, us that tray to make more trays, knock up your grains with those trays you've made and keep those trays going until you've got all the grains you'll need knocked up. If done like that, the slant should go bad before you've had a chance to use all of it.

Take a piece from the master slant transfer to a new dish let it grow out transfer that again now you have plenty of clean mycelium and your master slant is still in tact

like the guys above said

what i do is use master slants for long term storage in a dedicated fridge, then when i want to work with that culture i drag a loop through it (basically just scrape off some tissue) and return the slant to the fridge, and place that tissue in the center of a plate and allow that plate to colonize. when that plate has a healthy culture, i make LI from that culture, and use that to inoc as many jars/bags as i need

alternatively, you could g2g and expand it even more before spawning to bulk
