Ziploc bags are not air tight, you can suck the air out and zip/lock em up and the next day they lost vacuum. So they are absorbing moisture from the air. Vaccuum sealer is the way to go :thumbup:

Mainly to ensure its clean.  Doesnt hurt to go 2 or 3 transfers before inoculation.

I would slant for easy access to fresh genetics unless you want to start over from ms.




So I f I don't have slants couldn't I just store a culture on a regular dish?


what are the benefits of slants?

less surface area, smaller footprint in fridge too. less wide opening means less chance of rogue contamination during sample retrievals

The wood involved in making culture slants aids in the long term storage of mycelium in culture slants too.

Hello.
I have a question about Pan Goliath. I made one cacke from horse manure and coco fiber. Then i did caysing mixet up with another portion of manure and little bit of coco fiber and little bit of straw. When mycelium started to go out from top layer,i put tat cake to the plastic bin. with tepmerature 23-25C and Aeration every 2-3 hours.Light 12-12 from the window. Seemd that mycelium grew vegetatively. I been waiting 14 days and didnt get any pins,after that i gave him new top layer and put back for new incubation.
Right now i got 5 more cakes they are on the 7th day and seemed same story,but yeustarday grew one mushroom from the very bottom of the cake where is more humidity(i think).I heard the like more air and less humidity.
But all other cakes just grow vegetativeky again.
Can you help me please,what wrong I do?
What they do not have enough?
I grow Panaeolus for first time and really wanna try them grow better and try them too of course:)

The mycelium on the top of the cakes got too dry. 

What is needed is to do instead of putting a casing of bulk substrate materials on at full colonization is to put a casing layer of materials that the mycelium, or invaders won't want to injest.  Such as vermiculite/peat moss/hydrated lime/gypsum, and increase fresh air exchange.  Don't let that casing dry up either.  Mist lightly for the sole purpose of keeping the casing layer slightly damp.

Placing the casing layer on, then putting the substrate back into incubation, so the mycelium can colonize about a quarter of the casing layer would be ok too.

thank you for the quick response.Now i see how actually i have to do. Can I use only coco fiber for top layer instead vermiculit(its very hard to find it here) or other stuff that you suggested ?

You have to do what you can do when you can do it.  Never used coir as a casing cover for Pans, and would keep any, and all coir away from Pan cultures.  All that's known is that mycelium enjoys munching on coir, but peat moss?  Not so much.

Really? Typically mutants are denser in tissue mass. Especially blobs.

I do actually like how dense they are, I can fit a bunch in a small space.  And theyre potent, but they dont look like shrooms, which is important to some people

How old is the growth?
What species are you trying to grow?
What that mouth do?

So I have to get to some agar work yet tonight, but I kind of want to take a small dose. Normal for me is 3.5-4, I just want to take like half that at most. Been a while since I just did a low dose and vibe to some good tunes. I am just wondering how some of you long time grown feel about the idea of a low dose and then having to do some agar work, this for me would only be a lvl 1 trip, maybe just pushing a 2. Have any of you done it. Think I will do shitty transfers?
