White contam on agar?
    #24261804 - 04/21/17 01:06 PM (7 years, 8 months ago)

Hello, I'm new to the forums and mycology. I've been trying to only research and ask minimal silly questions but I think I've run into my first contamination.

I just started my first attempt at using agar using no pour (PP5) dishes with a poly-fill filter on top and pressure cooked @15psi for 90 minutes.

I inoculated the agar in a SAB with a spore syringe on 4/18 (3 days ago) and I'm already beginning to see some growth on multiple plates which I assume is bacteria. Could this be caused by the excess condensation in my containers or is that not something to stress about? Also if this indeed is bacteria should I throw away the containers or can I re-use them somehow?

If anyone also knows what kind of bacteria/contam this is I'd love to know just out of curiosity

If you had it under the scope, then maybe I might be able to have a stab at what genus/species it is, but it's bacteria for sure.

Thank you for clearing that up, guess I'll be making up some new plates. Buying a scope wouldn't be a bad idea for this hobby!

I'm still curious if having condensation in my plates is an issue and just asking for contaminates, or if the only issue it creates is a hard to view media?

Bacteria got in there because bacteria got in lol. Doesn't really just spawn from water.

Could have also been from the spores used, not uncommon to have bacteria in a spore print/syringe.

the general consensus is no... that said people dont like cultures growing to the edge for this reason... I read along time ago to just ignore it, I do... the shit pisses me off sometimes but my plates are clean so thats what matters

I use pasty plates and work with them upside down, it works great, try it out...

Thanks for answering my questions guys, really helps with the guess work.

I'm guessing the bacteria came from the syringe because this plate was a bit of an experiment where I used a few drops from the syringe which I'm aware is not recommended. But hey I'm the kind of person who needs to touch the stove to realize it's hot haha.

When you're working with the plates upside down are you just talking about when you're inoculating in the SAB or do you let them colonize upside down?
