From Multispore to Isolate Question.

What's the best way to go about this?
What would be ideal for me is to put a MS solution to agar, get a good culture, and make liquid inoculant following how frank gets shit done, and put that to grain then spawn bulk to CVG in 105qt mono's.
Now i have played with agar before about a year ago, but i was never successful with it.
MY QUESTION: Would it be a bad idea to make an isolate by doing sector transfers on agar from the multispore? The reason i question this is because i wonder the chance of isolating bad genetics and putting it to a bunch of grain, wasting a lot of time and supplies for me. Should I just make a few transfers until I have a healthy looking plate and go with that, then make isolates from the grow results??
If it sounds like i don't know what I'm talking about, tell me.

You'd have to do multiple plates , and label and put to grain. test test test. Or grow it out and clone.

If you don't have cultures you're going to have to start with spores. Once you have isolates you'll have to grow them & see what's what. You won't know much about them until then.

Ok, so make isolates then grow out and test. I'm doing a few different tubs, so i'll test and label a few different plates.
Also, don't know why I referenced to MS solution in the first post, I'm using a print. Same difference though.

Liquid inoculant and liquid culture are for people that not only do agar work but are competent and successful with agar. LC and LI will rape you if you're not good at agar work
An isolate can take 10-20 transfers from your first plate. Not many of us give a shit about isolates since most of them will suck you have to test dozens to get a keeper. Rsther than Dick around with that we just do a MS grow then clone something and keep the clone culture around
Sometimes clones are isolates anyway

When you isolate strains the goal is to have many to test from, not just one.
Start with a germination plate, and make a transfer of clean, healthy growth. Keep repeating these transfers until the culture starts sector prominently (this can take up to 8 - 10 transfers easily). You then take a transfer from each sector to a blank plate. If you are lucky you will have some isolates but I usually need to make another transfer or two if they still show sectoring. I try to get at least 10 isolates from a culture. I store a piece of every isolate on a blank plate in the fridge and I label and fruit out each culture to test
