What.
    #24256316 - 04/19/17 06:57 AM (7 years, 8 months ago)

So my agar was a complete failure.

2 days ago I spotted what appeared to healthy myc growth, just now I noticed that the growth was infact a slightly brighter shade of green, i gave the GE hole a sniff and it smelt like rotting apples, all 3 of these plates had the same trates, trich and some other weird contam. My B+ plates also have no myc growth and just this weird stream of pale bubble like mold inside. And theres nothing in any of my GT pots either.

Now I followed the LMEA agar recipe to the letter and pressure cooked a ottle full with polyfill in the lid, sterlized my pots with alcohol and poured in the agar, all this was done in a SAB on the 11th with extremeley sterile conditions.

So wtf has gone on? Is my house really that dirty?

Pics:

E plates (mold - no myc growth)

Try again?

Get some sterile petri dishes, some parafilm, or saran wrap...follow a tek.

Also in your post, include what tek you used, more specifics on your inoc. procedure etc.

Not enough here to help you.

Here is your problem. Alcohol doesn't not sterilize, it only sanitizes. This is a great example of that.

When we pour plates from the sleeve they are sterile.

With containers like these, we don't pour. Well not unless you sterilize the containers in the PC first but that would be a pain.

Mix up your agar, (I mix mine and heat it in the PC or microwave until its all mixed up like its ready to pour).

Then pour your plates, wrap in foil and sterilize in the PC at 15PSI for 45 min.
Bring the PC up to pressure very slowly if you get condensation problems, and if water is getting they the filter fold a paper towel and place it over the lids before wrapping with foil.

Yea can't use them then.

You can get 8oz microwavable deli containers (search for that online) its basically what you have but PP5 code plastic, PP5 can go thru PC cycle.
Most of us use Glad Mini Rounds like in my photos here, search Pasty Plates.
You can also use 4 oz mason jars.

Glass plates kinda suck. They're heavy and clumsy and expensive.
Buy a case (or a few sleeves) of disposable pitre fishes

Yea man! Those are sweet I'm ordering a bunch of those myself. The Glad Mini Rounds are fine but you can get 100 deli containers for like 5-10$, and they have a clear Lid. Wider too which is awesome

Instead of polyfil try a double layer of micropore tape over the holes. Its easier

nobody here uses glass dishes on a large scale. they are fucking expensive. those containers are way more
suited for no pour style. pouring is fun but its a vector. id just do those containers pasty style and call it a day.

you can use the saem agar. just PC them in the plates not that bottle and skip the pouring.

Alcohol is a sanitizer. Good example contaminated alcohol wipes killed a kid in Texas

Look up triad alcohol wipes out of Wisconsin. They didn't sterilize the starting material for making wipes since alcohol should take care of that crap anyway and low and behold contaminated alcohol wipes, dumbfucks

i was just telling my wife that if(or right now) we do have a 'third world war' it will be where the western system
starts to fail due to ignorance of science and lack of funding. and people will DIE because of it.

example. my mom had to call 911. they live in the middle of farm land. the EMS never showed up. they got lost.
i called the station screaming like a madman and they say, the new budget(pushed by guess who) had a cut in
the amount of 'subscriptions' to local and state gps uplinks.

basically the ambulance gps uplink software expired and it wasnt in the budget to renew the old one and nobody
bothered to source a replacement until AFTER it expired.

off topic i know but damn.

Why should gps be a paid subscription at this point? Thats SO early '00's. You know what really happened? They were dumbfucks, and wanted to blame something to not look dumb. Thats the price you pay living in bfe, servises may not get to you, and if they do it will be slow as fuck. Its like this in every nation tho bro. You think north korea even sends ambulances to the country side? Would that be a bike with someone blinking a flashlight?

I wonder how people ever lived without gps... i wonder if people hired drivers who knew the area and how to read a map and write down a address.

Im not trying to be rude to you, it sucks you needed help that didnt come, just sayin, kinda shoulda expected to have to drive in yourself. Having lived outside of towns a lot, thats what everyone i know would have done.

how do you drive when you are having a heart attack? so when those public services you PAY for fail you. you die
but its your fault because 'you should know better'?

you are being rude.

And honestly quite moronic. There's hospitals with ambulances in small Hick towns for a reason, which wouldn't exist if people drove themselves in.

attitude and cant read a fucking map or know there own local roads.
