Just out if curiosity Ham, what method do you use to collect spores?
I always put a little pin in the top of the cap, hold with one hand and decapitate with a scalpel in the other hand. Place through arm holes on a big sheet of clean foil that is already placed in my sab (I do multiples at a time). Let sit for a day, iso my gloved hands and arms and remove caps. Let sit for another day to dry.
Then i cut into squares and fold. I'm sure there is a better way, i particularly dont like cutting the foil afterwards. Hmmmm der maybe I should precut the squares....

I cut the foil first into 8x8 inch squares. Then when print is complete I fold it over in half. And fold on 3 sides. When you open the print it has a nice cover over it for protection to swipe spores ECT. I've got a diagram in my print method thread that shows this. I'll pm it to you later. Or dig in my references.

I used to cut 4x8 pieces, put cap to one half, and set a clean pint jar over it, while in SAB. Then let set 24 hours. Next day I removed cap and would stick a piece of wire under the very edge of the jar to allow air exchange. Easy as fuck to sterilize some thin jewelry wire with a lighter. After another 24 hours they would be dry. Fold foil in half and fold 3 sides.
Now I have been making sheets of prints in a Rubbermaid container in my SAB. The cutting apart of the prints does suck, but I soak my scissors in alcohol and them take the torch to the blades. I then tare a strip of foil about 8 inch by length of the box. Mist with alcohol and fold in half. I then set it in the sab and fold open some and wait for it to dry. Then cut the folded piece wide enough for the print and bent shut foil around edges. Slip print in and fold 3 sides. You do have to make DAMN SURE the foil is dry after alcohol spray. I get 4-5 prints from each bent over piece. You could just print on this, but I REALLY like having the print not on the "cover" foil, but that's a me thing.

can you autoclave that hole punch of just sanitize it? I'd want to keep the foil off the roll sterile you could do a stack of a few foils and throw out the top and bottom I suppose

this germ plate pleases me. keeps pushing out pins. ive cloned 3 so far. why not 4?

this one is ready for its first cuts. i got lots of good places. i could wait but the dish is sloppy.

and this guy too. ill be taking cuts from 2-5 o clock.

i nver thought to pc foil like that. not a bad idea. i always wipe off with iso but for giveaways ill be doing that.

In other relating leucestic golden teacher news; I made a 5 part + 2 part cake mix the other day. Had some grains, and cake mix left over, so I threw the grains inside of a spawn bag with the cake mix on top of it, and sterilized the bag for 3 hours at 20 psi's (All American), let it cool, and inoculated it with that LGT LC by free pouring half the contents of the jar (LC) into the bag. No shake, nothing special. Sealed up the bag as usual, and placed it on the shelf to colonize/incubate.

I'm pretty sure I'll experience something similar on some of my mini's. My wheat spawn had some metabs during the end of colonization and the myc looked stressed. Started out good though but.... I quit using rye and wheat because of this problem, either I'm prepping it incorrectly or pcing incorrectly, who knows. I've heard grain farmers treat rye and similar food grade grains with endospores to limit the liability of the grains growing a harmful mold if stored or prepped incorrectly by consumers, and maybe my pc can't kill these endospores. Some say a pc will kill anything, some (like me) think it actually doesn't. I can never see or smell colonies of bacteria but I see stalled growth and metabs, so I know it's there.
I moved on to pure whole white millet a while ago (birdfood, not a wbs mix) and I have never had a bacterial jar with that grain.
I'm willing to accept that I'm doing something wrong but I can't pinpoint what it could be. I have definitely seen a pattern with rye and similar food grade grains (sourced from a health food store). My rye prep is on point, I vent the pc for 10 minutes, and pc for 2 hours after the pc is stabilized. I'll just stick with millet.

they make strips with 1,000,000 bacterial endospores from much harder to kill species than ones found in grain and a regular old 15m at 15psi kills all of them. we just use longer times because we're sterilizing grains not liquids or instruments.
we don't run into Geobacillus stearothermophilus Bacillus atrophaeus etc.. we're getting clostridium for the most part.
they also make vials with 107 spores per mL so you can use 0.1mL and add it to agar then sterilize the agar and incubate dishes made with it to validate your autoclave and make sure that 15m at 15psi is working to sterilize 1L or smaller volumes of agar.
