Nice Everybody! This thread is going places. Keep up the good work! My spores fell on agar only yesterday.
Thinking about chasing an isolate, or maybe two of these genes for researching those questions/methods that can't be answered without using an isolate to test them. Should be fun.

  1st transfer underway. New LME recipe, second time pouring plates, I plan to transfer to at least 3 of the dishes pictured.

i can tell already this waiting part is going to kill me aha, all i can report right now is no contamination visible, but its only been two days so.. on a side note- since my grandma moved in with me shes had no need of her kitchen ware, was looking trough it and had to do a double take when i saw an oster blender then looked up and saw a weight to a pressure cooker, imma have to find that pressure cooker

Good looking culture, Germs. Looking very tomentose. I see 2-3 sorta sectory looking sections between 5-6 o'clock on that culture.

wasn't quite sure when i looked yesterday but now i am.. and i was right in guessing the brf plate Woot Woot!! 5/4 inoculation 5/7 clearly visible germination on brf plate
id put a photo up but we ll know what it looks like plus itd be an extra crappy photo because my phone keeps focusing on the glass for some reason -_-

Very cool Cepheus16. Put up a pic when you can. Also, try holding the camera further away from the plate, and if you can focus in from there.

Managed to take 3 transfers last night. Germination plate was poured thin which worked out good. I was easily able to slip the scalpel under the agar so I was able to flip them mycelium side down sandwiching the cultures. Hope this helps them get ahead of any bacteria that might be there, I had read some where about doing that.

Cool, Mad. On agar mycelium runs faster than bacteria, and mold, so another cool trick is to take your transfers before the culture runs into the walls of the plates, transferring away from the very leading edge of the culture with a wedge of culture cut no larger than a grain of rice. That way the mycelium has a chance to out run any contaminate that may be present. Once the culture hits the walls, any contaminate that may be hitching a ride will be able to catch up with the mycelium, and the whole culture becomes infected.

as per request- it just showed up yesterday and was barley visible. this is about as good as i can get the pic, partly(mostly) because of the jar im using, and my camera acting up.

also my first real attempt with agar, so excuse their(its) crude look.

I have started watching my plates daily. I have been taking transfers at about 1/3 - 1/2 way to the edge, seems to be working better than when I let them get bigger.

cool, ill keep that in mind. out of the 5 difrerent plates i have the brf is the only one any where near this far along, there is another that could possibly be germinating but its to soon to tell. also the brf is the least stiff/smooth of the agar.

Thanks! Dude, it looks good Cepheus. What are you talking about. You're excused too, because cultures are hard to photograph in those squat jars. I like the little arrow pointing to the young culture. Bravo.
