Have some agar plates in SAB

Hello all, I have some agar plates PC'd a week ago that I left in the SAB, are they still able to be inoculated?

depends on how you made and stored them...

were they wrapped in parafilm or in a plastic bag?

pics would help

typically we just use SAB for clean work, not storage. the SAB is never sterile, we just leverage gravity to our advantage, since contams accumulate on the horizontal surfaces

if these were pastyplates and they were airtight, they should be fine. really as long as they arent contamd they should be fine

i always use plates within 24 hrs, but some people keep em for weeks without issue

#1 at this point we have to assume the lids were put back on.

#2 if we assume they were wrapped in parafilm or cling wrap or put back in the plastic they came in they should be fine.

#3 if the lids were put back on but they weren't wrapped then they are still most likely fine.

#4 if you see growth on them and haven't put any spores on there then they are fo sho bad.

#5 if the lids were never put back on, then again they are fo sho bad.

ultimately tho if there isn't anything growing on them then you can use them and transfer out only mycelium and be ok. that's what they are for after all. i wouldnt just toss em because they were sitting closed up (wrapped or not). ymmv and good luck

I keep extra pasty plates wrapped up and ready to go, just look for visual contamination.

PC'd the plates with the lid on them. Had a wet paper towel on top and then aluminum foil on top of that. I removed from the PC and left those two in the corner, one stacked on top of the other. Noc'd three plates and set to the side of the SAB, got mycelium on one of three there. The other two, only contaminated : |

So is agar used to isolate specific strains within a spore sample? I never understood its purpose.

It could be , eventually. Most important it is used to make sure your innoc is clean. It later very easy to see any contams and if you do see contams, you can take a piece of good growth and transfer to a new plate to clean it up more. Bohdi has some good agar links and Stro also has a good one one cleaning and isolating. Search for those two for a good rundown

You're supposed to use a *dry* paper towel not wet. The purpose of the paper towel and foil is to keep things dry :lol:

You can use agar to isolate strains, you can also take a tissue biopsy from a fruit and clone.

By germinating spores on agar and transferring clean mycelium to a new plate, you can be sure you are working with a clean culture. This is what is most important about agar.

It is also used to store live cultures. A plate can be left in a fridge and then taken out as needed toinoculate grain spawn, having clean mycelium and skipping the germination times.

Growing from spores will give you different results every time while a live culture on agar like an isolated strain or clone will give you consistent results
