Haha for sure!  I've preped grains and plates so I'm comfortable with the pc, but defiantly a good point. Nothing like fighting with a jar in a SAB, while your mother fucking the world the whole time. I can't wait to give it a shot. I've read and seen a lot of good things about LI, so when i get these plates grown and transferred so i can give it a go.

I have a very simple rule for my LI plates. if its anything besides a perfect circle, I don't use it. its a bit overkill, as sometimes pieces dont come off the blades just right, but i just reserve those plates for transfers. ideally, i have 2 plates for each culture. one plate for transferring, one plate for dropping into LI.

anyone mess around with surface dryness?
as keep the sub wet with bottom watering but leave the surface slightly drier than normal?

well... the surface bruised and i cant notice much knotting.


the sides and bottom(nice and moist) have knots coming in nicely.

What you say it's a recipe for failure. It's true that water can be effectively sterilized just by boiling, but in order to keep it sterile it has to be kept in a sterile environment, obvious, no?

If you boil water in a pot and wait for it to cool in the pot, you can bet that water is no longer sterile, not to mention when you go and open the lid of the pot to suck up the water... you could just as well suck up water from a puddle in the street and call it sterile :thumbup:

The purpose of using distilled water to make spore syringes is to prevent spores from germinating in the syringe, in case you want to store the syringe for later use. The minerals in well or tap water encourage spore germination. Distilled water does nothing to help keep shit more sterile.

Regarding your earlier post, I'm not sure what you mean by a filtered container.  Is this a container with a SHIP?

I'm asking if it has a SHIP to stick the syringe into in addition to a filter.  Like if the container is to be opened in the SAB if it just has a filter, and if so what kind of filter and also what the purpose of the filter is in this case.

ships are for needles, if there's no ship you have to open in a SAB or in front of a flow hood

filters keep out contamination that's in the air

I still don't see the relevance of a filter here when you don't need gas exchange since it's just a container with water that is to be drawn into the syringe, unless it is for preventing a vacuum when opening in the SAB.

Also the implication that the boiled water in the pot would be such a great vector for a syringe in the scenario I described is lost on me the way Josex puts it.  The pot's interior will have been steamed and the water beneath the surface would be virtually sterile prior to opening the lid inasmuch as boiling water sterilizes it, which seems contrary to what I read in the PC manual regarding bacteria.  Anyway, sure, when you open the lid of the pot, contaminants will land on the surface of the water, but beneath the surface I don't see that contaminants would have infiltrated the water in the small time frame that you remove the lid, although withdrawing water beneath the surface could suck water from the surface into the syringe.  So I see the possibility of a contamination via that route, but not one as extraordinary as described.  Note that this is just speculation about a last minute method that could have been done to hasten a nearly finished SAB session.  Of course in general for salvaging a syringe for future use or creating new ones I would be inclined to use an SAB and other complimentary steps.

Anyway, regarding agar wedges, didn't you say that you would noc up several jars with one dish?  I have five decent looking clone transfers that I want to use to inoculate 10 rye jars from, but maybe save one for further transferring/storage, giving me maybe 4 dishes to work with.  I want to get some monos rolling from the clone.

And while I'm at it, regarding CVG bulk, is it common practice to just squeeze excess water with one's hand to get it to field capacity if it is too saturated?

It's odd that we don't have a forum on "cooking with mushrooms", not even for the gourmet mushrooms.  No recipes or anything.  Maybe I just didn't look hard enough

can I rehydrate dried cubes in warm water and then add to mushroom soup?  Or maybe rehydrate then pan fry in butter?  I tortured myself eating that plate of fresh cubes and next time I want it to be enjoyable.  I'm thinking Campbell's Everyday Gourmet Mushroom Harvest Soup would be a good base to add some rehydrated cubes to for a yummy treat.

But they taste so good when you chew them up dried.  At least when you're only chewing on .2 gram, maybe not so much when you stuff a handfull in your mouth. 

I bet I could make some pretty tasty dishes with cubes, I'm going to just pretend that potency isn't affected so that I can be as creative as I want with them and come up with some gourmet dish, maybe increase dose a bit so to balance potency loss. 

Mushrooms are so weird on the gut, when I'm tripping I can't figure out what food is anymore... I get this feeling eventually like I want to eat and then I look in the fridge and it's like... dead stuff... ewww        Then I start dreaming about mango nectar or some fancy elixer of goodness and I swear off all non hippy food for the rest of my life...

Then I sober up and eat half a dozen eggs and a pound of bacon.... it's a vicious cycle

skin-on fish is the only thing that doesnt have that 'dead' inanimate edge to it. but even veggies seem ...rotten.
