It's a lot of work to test isolates, especially when you test them full scale to see if they do well with your grow method

Yeah. I agree that it's a lot of work. But I think it might be worth it in the end. Just finding the "Knock your socks off" sub strain is a cool and rewarding idea. I'm about 65 plates into it with PE ms and just became curious when i saw that water running over the sectors. It seems to make sense that the water could carry little bits of myc and possibly do away with further isolation. just thought i'd ask.
    I also agree that a ms clone is totally easy, quick, and acceptable. I just thought i'd give a first generation pure isolate a try this time. It's a fun experiment while I have about 30 different ms and clone jars, and two bags colonizing now, to fruit and clone the keepers. Mostly from syringe to agar to grain but a couple ms syringe to grain.

  EDIT- oh yeah and a couple clone to agar to grain.

And yeah, I've checked out that pussy fart thread before. A lot of work indeed but I'd just test all the isolates out in little trays in a shotgun really quick instead of committing full monos.

Betcha my good clone is 98% as good as a stellar isolate.

Water on a plate is more of a worry for carrying around bacteria or spores from simple molds but I guess maybe just maybe it could hamper isolation effort on occasions.

If you want an isolate though go for it but i definitely would get a limited MS grow and a clone going first

i bet you have a very sexy clone. the pictures in your tag (signature or whatever you call it) are beautiful. I have a couple clones going at the moment and am super excited to compare. I actually tried my hand at cloning a 6 month old dried dick and had great success as well. I was excited about that. thanks again man.

RR posted from ideal standpoints. Ideally it's even better to obtain 2 dozen monokarons, combine pairs until a great strain emerges.

Excellent clones can be found and maintained for years. Most people go that route because it tends to produce decent yields while you are looking. Isolation on agar is worthwhile but takes forever.

This is how I treat it. I mostly clone but I work on strain isolation in my free time. I really enjoy culture work so its fun for me

It is true to get single strain isolates would be extremely more easily done with single spore germination and then mating. A micro manipulation scope could accomplish that

Hey fellas.

  Do you feel like it's possible for water in a petri to leave stains where it ran without being contaminated? I've noticed very light, hardly visible stains from time to time that don't seem like a problem and don't seem to ride along when i transfer to new plates. I know pictures would help but I can't pull that off right now.

Could possibly be minerals in the water that are drying up, like water spots on dishes washed in tap water with high hardness. Like you said it's hard to tell without a picture though

I'll try and get some pictures up soon. All my petris from that last round are sealed up and in my myco fridge. I like to keep them on hand as backups until the next transfers are Obviously healthy and wonderful. They are a bit tricky to photograph because when i take them out of the fridge they condensate pretty badly.

I'm pretty sure its the hard water issue though. I'll do my next batch with distilled and see if it changes anything.
