Shomann said: Hey guys, up until recently I've never had any bacterial problems while working with agar.. none I couldn't fix on my own.
I've recieved quite a few prints I've been trying to germinate and I just can't seem to get them to germinate without ugly bacteria beating them to the punch at the innoc point. I've read that bacterial problems are normally user error, however I only have problems with some of the prints I have.
Any pointers? Tips? I'm fighting to have these cultures and I feel I'm at a dead end. 3 separate agar batches have been a dud. Thanks mushcult.
Oh and %.
If it is loaded with bacteria, it is possible to mix up a batch of agar with some peroxide in the agar. Not sure on mixing ratios, its like antibiotics, a last resort and only used to germinate the spore and get a clean transfer.
I have a couple syringes from NON sponsors that grew cakes, but are loaded with bacteria. Like bacteria blooms 1st and I get small little blotches of mycelium surrounded by bacteria. Luckily I ordered some antibiotics which I am going to use to clean some up soon.

The "grain petri" works great.
Can I do this with a print if I'm having germination issues? Bacterial print issues?

Peroxide won't do a thing in the agar. You may as well be adding water. Sandwiching the bacteria has worked for me in the past You could also try a warm pour
Josex was doing pf plates to germinate stubborn spores. He added spent cofree grounds to the mix which I think is a nice touch. Honestly tho moab I think you just need a little more patients. That syringe should germ on agar no problems

I'm hopeful I can get some of these PE spores to germinate, been trying on and off for over a year from a swab. I also snagged a small ropey strand to transfer from an over grown plate, a clone from a thick and dense mushroom from a cluster and put some a wild Texan spores to agar.. Can't wait to have some nice assortment of cultures I also just received a wedge in the mail.. atl#7 let the fun begin.

i have found some really cool textbooks on various myc topics over the years, mainly tracking down stuff people are looking for. i found some really great stuff this week, id love to share it most are DRM protected from databases and archives and shit
i removed the drm (duh) but dont want to break any rules by posting them.. id post em on public trackers but i doubt theyd get much interest
there has been some really great stuff published in recent years, very dense reading, but fascinating stuff

Is it normal for spores from a B+ print to have a rusty color instead of black? Regular cubes should have black spores, right? I have it on agar right now, but I can't be sure it's B+ till I fruit it.

Hey so I feel like I'm over thinking this
If I take a jar I believe to be contamed and throw a grain on a petri the bacteria should show correct.

If the petri dish supports the growth of that bacteria yes.

I know nothing in the field of bacteria. Should I make up multiple dishes to see if something shows?
I feel like putting grain to petri would give endospores a good environment to live idk
Or should I just get that microscope you recommend.

A scope is better.
Not every petri dish media will support or grow every bacteria.
Plate count agar grows a lot of bacteria well but still no where near all bacteria

I forgot to put foil on my WBS jars in the PC and it got up to 8 psi before I removed it from heat. Should I let cool, put foil on and start PC cycle up again, or should I just put it back on the heat right now without foil?
