Flow hood help

I just got my flow hood and I did the lighter test and 6 inches apart and the flame blows away  and I have a slight dead spot. What can I do!?!? Should I be worried?

Ask the manufacturer what's up if you didn't build it yourself

Otherwise if you did it yourself try adding prefilter or throttle the blower

I added silicone in top edge. Air was leaking out. I added a prefilter and it seems to work out. No dead space. But arrownd the filter edges there is some turbulence when I do the lighter test.


Soon I will cover the gasket to make it more pretty

that's because it's a flow cabinet not a flow hood. there's going to be turbulence at the edges as the laminar flow and relatively still room air interact

OK. So I shouldn't worry . I will buy the 1900 filter from filtrete. And do agar Petri dish test.

just stay within 2 or 3 inches of the edge and I think yoou should be fine.  some tape might help you remember

how did you mount that filter? im a bit confused by it... do you have it glued in or something? is there no trim sandwiching the filter against a furring strip?

Of course. Just has to be more careful and mindful

Running it first and having it in a room with gloss walls or wipeable surfaces and a hard floor that csn be swept and mopped are more critical with using a flow cabinet so the filter can scrub the room.

With a flow hood you can get away with a less cleanable room but still should not be put in dirty room.

Still air box works in my living room with two cats and some litter boxes

Awesome, my university has laboratory grades safety cabinets and I was going to do my work in their today. Haha you've got 2 cats and have no problems because of it? I've also got 2 cats and was wondering if that was contributing to the contaminations.  Good to know I can still make it work if I'm extra careful.

You don't think so?? My friend is graduate research assistant and has keys to her own lab with the flow cabinet. She says I can use it if I provide her with fruits later.

Sort of, but not exactly.  With a sterilize lab you are still going to need to maintain sterile equilibrium of the lab by cleaning all surfaces in the lab at least 2 times per month. 

You are correct in that you don't want any linen, or carpet in the lab, because they harbor unbenificial spore loads.  An easy fix to turn a carpeted room into a sterile lab without removing the carpet is to run plastic sheeting along the floor. 

All the maintenance involved with keeping a sterilize lab is worth the effort, because you have free range of motion during sterile technique which can become very important if you are practicing this hobby on any kind of "commercial" scale.

Don't bring a dirty mop head, or dirty broom into the lab either.  Brooms kick up dust, and dirty mop heads are a bacteria haven.  Paul Stamets uses a wet/dry vacuum cleaner that is connected outside of the lab for clean up of spilled grains, or whatever, and he uses a fresh mop head for the floors then disposes of it.  He also recommends that if you have a spill to stop what you are doing, and get the spillage cleaned up before continuing which is what I do too.

The poor man's way to achieve that is by manually wiping every surface of the lab with alcohol as soon as spills happen.  During the maintence regimen, I wipe walls with with soap, and water, and use rubbing alcohol on the floors, and shelving systems.

Soap and water to wash a lab to me means that it was too dirty to begin with.

I bleach all surfaces, wipe down, then bleach again and leave...Alcohol is used to clean surfaces before and after sterile work. CDC guidelines...

Nope, no battle here.

You're soap, and water theory is specualtion.  In both TMC, and GG&MM Paul Stamets writes about maintaining sterile equilibrium in the lab using soap, and water. 

Been running a lab in an extra room for 5 years.  I used bleach before, and it's too harsh for me.  Plus a lab, once set up, stays pretty dust free, so bleach is overkill ime. 

Do you clean your lab with bleach every 2 weeks, and how does that work for you?
