Good let it cool slowly. Yup some sediment is usual. Swirl it around when it's still decently hot and it should go back in suspension. Pour when your agar gets at or just below 120 F.

No, it doesn't matter what fruit you take the spores from spores are spores.
When you pour, wait until it is cool to the point it might start to solidify if you let it go any longer. This will prevent condensation in the plates , its the best trick I learned. Before I figured that out I always had condensation on the top couple plates, would have to set a mug of hot water on top of the stack as it cooled to stop it.

Sounds like you let it cool too much...I've had that happen... Pasty's easy agar tek is a little easier than pouring plates... Either way in my experience takes about the same time...I have glass Petri's and cleaning them can be a pain in the ass...then not to mention oven sterilizing them...think I'm going back to pasty plates very soon actually

I've heard that about taking a clone sample- reasoning being there is still a lot of "young" energy in the mycelium, much like transferring from the edge of growth on a plate. There are two problems with this theory. 1st is smaller fruits are harder to get a clean culture from, the second is I've never seen any data to back that theory up. As far as spores go it does not matter which caps you print- other than the size of print you want.

can someone help me out. i dont have a picture at the moment, but when i am doing spores from prints on agar the mecelium is very light. like transparent or what ever. what do i need to do different. i have put some of this agar to wbs and it grows out just fine, just curious as to why its see through and not white. once i put it in wbs it grows out a very briliant white. thanks
