so i just received some agar and petri dishes. now i want to make the agar and start learnign how to use it. my question is... my PC is not big enough to use a wiskey bottle or anything like that. i need to get a bigger one but for now it is what it is. can i use a pint jar to make agar in. just use less amounts. or can i just boil the agar mix in the wiskey bottle for and hour or so?

You can use a pint jar you just wouldn't be able to make that many plates.

Hopefully by agar you mean... Agar plus a nutrient such as Light malt extract I am not sure AGAR alone will work!

and i have sterile petri dishes i just use a bottle neck bottle. its to small for this little pc. so my main question is can i just mix the right amounts of malt agar and water for a pint jar. mix. boil. pc for 30min. and pour from the jar into petri dishes. the jar would have a ship lid on it so it already has a hole for GE

I pour all my petris from a quart jar so yes.
250mls will do roughly 10 plates.

You might find it at a health food place...but Walmart is not going to have it...an Asian store or online is the best bet though

You dont need to buy anything else , what you have has agar n nutes premixed. A mickey or mini mickey of absolute vodka works great for a short small necked agar bottle also

Check out media bottles. No need for a new PC. They come in different sizes. You can find one that fits your PC.

Just use the MEA, don't need Agar agar with MEA. 25 grams MEA to 500 ml water. You can scale it from there to whatever amount you want to make.

OK so I mixed my agar and PC it for 30 min. When I took it out I noticed there where some chunks floating around. I know it's not from anything else. Is that normal. And it's a yellowish color. Not clear

Good let it cool slowly. Yup some sediment is usual. Swirl it around when it's still decently hot and it should go back in suspension. Pour when your agar gets at or just below 120 F.

No, it doesn't matter what fruit you take the spores from spores are spores.
When you pour, wait until it is cool to the point it might start to solidify if you let it go any longer. This will prevent condensation in the plates , its the best trick I learned. Before I figured that out I always had condensation on the top couple plates, would have to set a mug of hot water on top of the stack as it cooled to stop it.

Sounds like you let it cool too much...I've had that happen... Pasty's easy agar tek is a little easier than pouring plates... Either way in my experience takes about the same time...I have glass Petri's and cleaning them can be a pain in the ass...then not to mention oven sterilizing them...think I'm going back to pasty plates very soon actually

I've heard that about taking a clone sample- reasoning being there is still a lot of "young" energy in the mycelium, much like transferring from the edge of growth on a plate. There are two problems with this theory. 1st is smaller fruits are harder to get a clean culture from, the second is I've never seen any data to back that theory up. As far as spores go it does not matter which caps you print- other than the size of print you want.

can someone help me out. i dont have a picture at the moment, but when i am doing spores from prints on agar the mecelium is very light. like transparent or what ever. what do i need to do different. i have put some of this agar to wbs and it grows out just fine, just curious as to why its see through and not white. once i put it in wbs it grows out a very briliant white. thanks
