LC lid with polyfill only?

I want to make a lid to use with an LC jar, but I don't want to deal with vacuum related issues and I won't be able to buy whatman syringe filters for a while.  So, my question is, would a hole filled tightly with polyfill do?

Can I shake the LC jar safely with a lid like that?

you could but you don't want to get the filter wet. how are you going to make the LC?

You don't need to shake, only swirl gently to break the mycelium with anything proper, get creative. You can use marbles, glass shards or whatever works. I've been using blender balls, they're awesome for the job.

Of course, a magnetic stirrer at max power will break the mycelium very easily in just a couple of minutes using a magnetic stir bar.

Polyfill sounds like the worst option possible and I think 4 or 5 layers of micropore tape would be much better, although it would still be far from ideal.

Whatman > SFD > MP tape > tyvek > polyfill

The filter should/may have contams on it if it is doing it's job. poly fill filters don't stop liquid from escaping the jar when shaken. It would have to be ungodly tight, and then you would be restricting GE. So if you shake the hell out of the jar chances are you are going to contam your LC.

Really? Micropore tape would be better than polyfill?  Gues I'll do it with micropore tape and swirl with something placed in it until I get whatmans and make a magnetic stirrer.

PTFE SFD > cellulose SFD > polyfil > real tyvek > post office tyvek > ez felt > mp tape

For a LC use syringe filters or be smart enough to use a different filter and not get it wet

Pfft if ya got plastic lids you probably don't even need a filter

Also I highly doubt a stuffed tight AF poly filter would have no gas exchange. LC and agar colonization is so small, it shouldn't even need gas exchange. I've grown many a dishes with literally no gas exchange holes, until you get into larger substrates, you won't see the myc stall out with such a small amount of media

But I do need holes if I don't want to deal with vacuum right?  and I think I need to have filters for those holes.  My lids are metal but what would be the difference if they were plastic?

With metal you don't have a vacuum if you have the lid upside-down or the rubber seal removed. In fact I'd recommend that anyways, nothing worse than struggling to get the lid off and potentially tossing in a ton of contams.

Plastic lids would be done exactly like pasty's tek. Half turned open during colonization, and then you close it tight before shaking

I use tall jars and regular grain jar lids. :shrug: maybe you don't really need to do LC

I use a plastic lid and syringe filters. Works pretty well and the syringe filters are autoclavable.

So I've finally bought some syringe filters and put them on lids using silicone.

and I'm following TL's ELME tek, which uses a gram of LME for 600 ml of water, which I did, and divided into three 400ml jars.



So what do you think, am I good to go?


I'm going to inoculate them with agar wedges and was wondering if I could just turn them upside down and stick the needle into the syringe filter and suck some LC when they're colonized?  Do syringe filters work like that?

and as I'm planning on building a magnetic stirrer with a computer fan and a magnet, would it work if I drop some screws in there (guess they could rust and I can find a better material) or should I just shake them as I already have syringe filters on them?

I use SFD's. I really like these Erlenmyer flasks with threaded caps/autoclavable lids, they're like 5$ if you find them on EBay or Amazon. I drill a hole and pop the SFD underneath.
The shape and height of the flasks means you can swirl without worrying about splashing the filter.

With a magnetic stirrer I think the little stir bar is a magnet.
You put it in before you sterilize your media.

I blend my agar wedge in water in a sterilized Eberbach first. I got mine cheap on EBay , but you can make your own with Oster blender replacement parts and Mason jars.
I then pour it into my LC broth and let it spin on the magnetic stirrer for a few days.

This is the best way to do LC IMO.

I wouldn't do that no. Just open the lid to pull LC- if you can't work with sterile containers open you might wanna re think using LC anyways

I already swirl it daily but it won't break and is getting bigger and bigger.  I think shaking shouldn't be a problem, considering the syringe filters are hydrophobic.  But I'll wait until someone experienced verifies that .)
