Poured my first plates today. Had a bitch of a time finding agar locally so off to Amazon I went. Got 500 grams of powdered agar for around $25 shipped.
It's killing me waiting on the plates to cool so I can start with the spore syringes I got from from a forum sponser Gives me a good reason to try the new gloves on my glove box.

Dunno. I'm new at this but everything gets the shit whipped off of it with Lysol and/alcohol. If it's badass enough to beat through that then it can have the damn plate.
Until I get a flow hood built, it'll have to do.

you only need water and a drop of dishsoap in a SAB. Lysol is pointless and alcohol will make it perform worse and make it dangerous as shit. a SAB that has been saturated with alcohol is a bomb waiting to blow up in your face, literally. we have had users goto the hospital because their SAB blew up from using alcohol on it.
It is fine to use alcohol-soaked paper towels inside your sab to clean plates, and you can use it to clean the box between sessions. but the ONLY way to prepare a SAB is to apply soapy water to the walls and LEAVE IT. the walls should always be wet with soapy water, not lysol, and definitely not alchol.

I would agree if it were a properly made glovebox but anything made in a tote will add unnecessary drafts

i actually misread that. i thought he was saying gloveboxes are better than sabs. my bad. been a long day.
a lab glovebox is obv the best. but in amatuer mycology, you cannot build a homemade glovebox that will be as effective as a sab.

people build flowhoods it's possible it just depends on how you do it. I wouldn't recommend it to anyone though so the point is moot

so I got into agar about a month ago thanks to pasty plates, I have a few clones on agar, these are 3rd transfer,

do they look alright, I was thinking of looping a few pieces to new plates then tiger dropping whats left to a few rye quarts in a day or 2, should I just do it now or should I wait until the plate is almost fully conlonized. any imput would be much appreciated thanks in advance.

iso soaked paper towel for wiping plates is all I use now.
I helped build a bunch of cleanrooms for a few months and I have to say they are amazing. The entire ceiling had the standard 2 by 4 grid but instead of tiles each cell had a hepa filter and dedicated blower. Can you imagine an entire ceiling made of hepa filters? Slits in the bottom of the walls allowed the air to escape and created positive pressure......what I would give to own one of those rooms.

Ive tried this like 5 times now without success.
1. For some reason this recipe is only making me 5 plates?
2. Nothing seems to grow on them. Ive tried tissue samples and drop of LC and neither showed any visible changes
Any help? Sure would hate for this big ass bag of oats to go to waste.

Why would oats be going to waste if this is a thread about agar? If you're only able to make 5 plates, it sounds like too much liquid is evaporating and it's becoming extremely stiff/denser than optimal. A good trick is to follow the recipe here by Munchauzen and for that recipe specifically notice he says "Upon boiling, immediately remove and pour into a measuring cup. You can use the the cup to see how close to 1/2 cup you are. if you cooked off too much water, you can simply add some back in."
