How do these Pan cyan and galindoii jars look?
    #24041265 - 01/26/17 01:14 PM (7 years, 28 days ago)

Hey guys,

I couldn't get some spore syringes to start on agar so I inoculated some small jars of wheat some time ago -while I was working on my flowhood.
Now the flowhood is ready, and I'm ready to make drop some grains on agar plates to obtain clean culture.  I just wanted to make sure what I'm transferring is what I'm intending to transfer.

I know these look pretty bad but keep in mind that all I'm trying to do here is to clean the culture.

Here is a jar of Pan. Cyan. Hawaiian, it obviously has bacterial contamination, and there are some yellow liquids coming out from the mycelium, are they metabolites from heavy bacterial contamination?

and this is a jar of P. Galindoii, it looks pretty fucked up but there are already some sclerotia formed (the big one in the pictures is lifting the whole substrate from the bottom of the jar lol), so at least I'm sure there is some galindoii mycelium in there.

Since I'm not familiar with how Pan Cyan or Galindoii mycelium should look, I'm not sure if there is ONLY galindoii or pan cyan mycelium in there (I mean other than bacteria).

It would suck to cut a grain from the white are and later on find out that I transferred some kind of white mold.  I'd really appreciate some opinions.

Almost forgot to mention that I have self-healing injection ports (holes with silicone on top of them) on those jars, which I used to inoculate (and did it in a SAB). I also wiped the jars with 70% alcohol very well and made sure the needle got glowing red between jars.  So I think the syringes' itselves were not 100% clean.  That's why we do agar, right?

Yup, got similar mistakes at the beginning.
Now, after swapping to Agar, the success is incredible.

 nice to see ya Tira.

Yeah the reason we use agar is also to tell the difference between molds and myc, the only way to know for sure with these guys is to take some myc and transfer.

Personally I'd harvest the sclerotia and take a tissue sample to clone, could try some grains too, mexicanas get VERY fluffy so it's probably myc on top there, and xfer a few grains from the pans as well

Thanks man, I'll make transfers from all around the jar and keep an eye on the plates then.
