I cut off a portion of the swab and streak the he'll out of it on the plate and sometimes I even leave a piece of the cotton on the dish (I also had a hard time germinating from swabs)

That's what I did.  I streaked 2 plates.  I didn't know if I can keep the swab now for later or not.?

You probably shouldn't put the cotton onto nutrient rich media if you plan on using it again :/

That's why I cut off a portion of the cotton or throw away the swab after its spent.

Yeah I kind of figured that.  I reluctantly threw it away.  I rubbed the shit out of it.  Hopefully it takes.  I'll try cutting it next time.
  How do you streak it after you cut it off?

I was reading HERE fishermansjc digs a little divot into the agar and cuts & drops the piece into the divot.
I may go ahead and try the cut method.

I move  it around with the loop or with a scalpel. Loop is much easier.

Why don't y'all just flame sterilize a loop and swipe the damn swab? No cutting, no dropping, nothing.

I haven't flamed anything in like a year or so lol I have a bunch of scalpels and loops so I just sterilise them all together with the other 15min PC items. Never needed more than 7-8 scalpels per SAB  session :eatingout:

The standard X-acto knife can fit into the common thin, glass culture slants that RR has in his videos. I dropped 10 scalpels into them and sterilized them in quart jars, 5 blades each. Now I have fully sterile blades, including handle, to dig into my slants without fear of fucking them up.

Just started a special shoebox of PCS from the wild prints I took this summer.  I actually combined the culture from my print with the culture from a clone in the same jar.  One was fluffy and the other looked like Doc’s hair from Back to the Future, lol.  Since they are the same species they seemed to combine and colonized my GS in 6 days.

I’ve done two or three transfers with this and I haven’t heard any crunching.  Doesn’t smell like mold either.  Smell is very sweet like the Caerulescens fruit.  Asked Bod a few days ago about it and was told it’s looking good.  I’ve posted some more pics on his Q&A page but haven’t got a response yet.

There is a linear pattern on the surface of the agar it’s just hard to make out.  Also if this was/is mold shouldn’t it have taken less time to colonize?
