First Agar Transfer Ever! *PICS*

Ok. So this is my second time trying to get something viable out of and old spore syringe to clean it up and get something of a culture going to put to rye berries.. The first time was an absolute disaster. Yes even compared to this.. lol As you will be able to tell I am still struggling to pour plates that don't condensate. I guess I'm too worried that the Agar is going to cool and thicken up and I jump the gun on pouring them. I'm trying to work on that. Anyways..
All plates are made with Agar Agar powder (from my local brewing supply store), LME, and water. (The new ones I added some food coloring to).
The Mess:....
I made transfers where ever I could see white rhizomorphic growth that wasn't surround by the funk:....
This is what I've ended up with.
First Transfer:
(Day 1)
So that's my first attempt at transfers. Hopefully something good comes of this. If not I guess I'll start over.
Thanks everyone for giving giving me the courage to even try this in the first place. Hopefully I'll only get better and better with Agar and I won't have to deal with this in the future.
...and I'll try to take better picures.lol
Sincerely,
BLiNK

I just had to move on for my own good. I love you all.

All I know is that this is the way to do it. I've had plenty of successful grows from multispore and plenty that were so-so. Everyone that has major success learns Agar. So ya know.. why not take the next step. Hopefully in a week or so I can post some positive progress shots of something much cleaner.

Cool good luck! Those first plates were the funkiest looking Agar plates I have seen yet. How do you wrap the plates? With Saran Wrap? Also, if you stack all the plates high after pouring that is supposed to remove condensation.

I'm just starting with agar as well. I was wondering about sourcing more agar and you said you got yours from a brewing supplier?
Just curious, it'd be nice to not make the drive for the stuff I got before if I can avoid it.

Sometimes health food stores have it. It's a vegan substitute thickener. It's used in some European and Vietnamese cuisine too, if that helps any.

If you have heavy bacterial contamination, you could try a hot pour. that's where you pour sterilized hot agar on top of the agar. mycelium should be able to grow through the agar whereas the "pasteurized" bacteria are knocked back. As soon as you see some white growth peeping through, just scrape it off with a scalpel and inoculate a fresh plate.

do you think this would help with picking strong genetics?

Try Ferather's antibacterial agar, just brew some black tea in your water before you put in any agar. Grows slower but I don't run into comtams this way

I use GLAD Wrap to wrap them. That seems to be the main recommendation unless you can afford to spend some money on parafilm.
I have stacked my plated everytime. Even pour them while stacked but I still get condensation. I've been waiting longer and longer to pour the Agar each time and I'm getting better and better results. This run being the best so far.
I also feel like stacking the plates is more effective when you have more than 3 or 5 in the stack. I've been doing small runs between 4 and 8 till I get the hang of it.

You are doing the "pour" method correct?
Bodhisatta mentioned in a post about pouring at 47 degrees I believe.

Thats right! I start pouring at 117f and get little condensation, which goes away after a couple days

How would I tell the temperature of the Agar inside the bottle? I've heard of putting the bottle in hot water to cool after the PC run but other than that I don't know how I would be able to tell. I obviously wouldn't want to pop the cap off and shove a thermometer in it.

I believe he placed it in a bath of water with a thermometer. I don't know the details, I just remember seeing something.

Yep. If you have one around I'd suggest you give them a call. The guy as the store even asked me if it was for mushroom cultures. He said it's a pretty common item customers come in for. Even said they stock petri dishes if I need them as well. <3

It's also a vegan thickener for soups and stews, as a substitute for gelatin.

Its an infrared thermometer with a laser pointer. You can get it at home depot for 30 bucks. Works like a charm!
