Not sure if flowhood is working right.

I've finished building my 24x24 flowhood and tested it with 15 agar plates. I first tested the flow with a lighter, when held against the flow, it bends about 60 degrees without flickering, so I think that's good to go.
Then I placed 5 plates and left them open for 30 minutes.
After that I've done the same with 5 more plates, this time 5 minutes.
And finally I've left open 5 more plates for a few seconds.
The thing is, there are plates showing growth in all three groups, so there's a problem to be fixed. Is it possible that;
-the flowhood's not properly working?
-I didn't make an extra effort to clean the room before doing this experiment (I clean it almost every morning anyway) and this might've caused the contamination?
-sterile technique fault? (though I didn't put my hands or any other thing between the open plates and the flowhood.)
-other?

You need to leave the flowhood running for at least 1hour before doing any kind of sterile work to ensure sterile air is being pumped through.
I am not sure about the 60degree angle but I think you're a little off so adjust your pre-filter accordingly and run a couple of light tests until that flame is at a 45 degree angle
You don't need to be spotless everytime when doing laminar work. Just wipe the surfaces with iso, flame sterilize your scalpels, needles...etc and wipe your jars/bags/whatever, wipe your hands with ISO frequently, always use gloves, a face mask won't hurt, wear clean clothes....etc
I fully clean my lab twice a week sometimes only once and I have had clean spawn with no problems

Maybe that is the problem, I only ran it for about 30-40 minutes before testing the first plates. But it was working for more than an hour when I tested the last two groups of plates.
Would opening the door briefly for getting in and out of the room cause problems?
I'll try adjusting the flow, I have a speed controller connected to the fan, so that won't be a problem.

Unless you're getting in and out of the room every 5minutes I do not see a problem.
My filter has been running for 3 days straight right now and I go in the room frequently. Sometimes I had to take desperately needed shits in between work and I've been fine just make sure your hands are clean, surface areas are clean, tools you'll be working with clean...etc
You should also think about the position of the filter.
My door is on one side of the wall breadth wise so I placed my filter along the length of the room, away from the air flow direction of the door so when I open the door it isn't directly affected. It's just a precaution that you might want to re-consider

OP-what kind of filter is it? are you positive its secured into the box, either with silicone or dry fitted w/ weather stripping? sometimes if the box isn't sealed, dirty air leaks thru the cracks around the filter face

I think RR quotes btw 45-60. I prefer a little stronger too though-almost 90.
its weird all the plates got contamination. I would think a slow flow would at least protect some in the middle
I open my door (and window is always 2-3" open) when working in front of hood too, so I don't think its that either.

Maybe not sealed like you said or not enough cfms

Do you shower before each session infront of your FH? And Lol on your video you couldve held the lighter the other way and you wouldn't have been roasting your skin

My suggestion is always a SAB unless you have a room without carpet and high gloss painted surfaces. I just personally think a flow hood in a regular house room is a pretty shit idea/plan. I know people do but I wouldn't ever waste my money on a FH just to be put in regular room

Sometimes I do, sometimes I don't. It all depends, if I showered the night before and haven't really done anything, went to bed and woke up the next day I generally just enter, roll up my sleves, put on some gloves and a facemask, wipe down with iso...etc
If however I've been at UNI, where I study Agriculture so generally a lot of field work or I've been spending a day messing with my 4x4 then yea, I generally make sure I shower and put on clean clothes before
And yes I know I could have pointed it the other way, I get ripped on about that alot

I actually have a dedicated room where I placed my FH and all it has is tiling on the floor and the walls are bare. The rest is just a small shelving unit for my work so alcohol lamp, boxes of sterile gloves, packs of paper towels, ISO 15L containers, sealed petri dishes...etc

I secured it to the box with silicone both from inside and outside of the box.

I also read RR quoted between 45-60, they didn't all get contaminated btw, just 5 of 15 plates but I think that's pretty high for a flowhood.
I actually expected the ones I left for 30 minutes to contaminate, and even some of the ones I left 5 minutes, but it's weird that the ones I briefly opened, held in my hand and closed to get contaminated. I think I fucked up somewhere with the sterility and it's probably not the flowhood's fault I'll try running the experiment with better conditions.

I'm currently trying to get into bulk oyster production and the reason I've built this flowhood is that I want to work with sterilized bags a lot (also lots of a agar work)
