Rye Berry w/ Aspergillus at inoculation sites?
    #24019707 - 01/18/17 11:26 AM (7 years, 12 days ago)

Hello, it's been 7 days since inoculation and discolored (grey/black) growth at every injection site on my jars.  I used a spore syringe from a reputable company.  I flame sterilized, then wiped hot syringe w/ alcohol to cool off in between each jar.  Although I used a pot of Dark Roast Coffee w/ gypsum in the soak water, would the coffee change the color of mycelium?  Or did I somehow contaminate my needle with the alcohol paper towel used between jars?  Seems odd, because I used a new alcohol paper towel every 3 jars, prepping about 10 jars total.  All jars have identical growth at every single injection site.  After research, the color of this growth is leading me to believe it may be Aspergillus?  Any advice is more than welcome.  Thanks guys.

Did you follow basic sterility factors? It would seem as tho you did not. I am fairly new to this forum, but not to mycology. That is definitely contaminated, could be Aspergillus, could also be botrytis/grey mould, either way scrap the whole deal, are you PC/steam sterilizing your substrate and containers? As the contaminated areas are all at the injection sites I would consider your "reputable" supplier as being the culprit, or you injected open air, which is ill sdvised, glovebox or flowhoods are a must for the home mycologist, as a lab grade clean room is very costly. Dont fret, just start anew, set backs are part of the experience, but this time 100% sterility, still air inoculation, and tyvek, micropore tape etc, any other questions give us a shout.

I put the substrate in for about 2 1/2 hours of steam sterilization. Although I injected the jars without a glove box, or anything.  I'm guessing that was my problem.  Are these mason jars done for, or should I toss the substrate / clean the jars while outdoors?  Or should I just toss the batch as a whole?

Toss contents and clean. Next PC cycle will kill anything leftover.

You do not have to toss the jars, just the contents, and take 20-30mins and make yourself a suitable glovebox, very simple and will have a drastic impact on your cultivation success, if you need i have photos of two newly contstructed gloveboxes i made for some cohorts, and there are a bunch here and elsewhere, dont be disgruntled, just clean it up a bit, and sterility, sterility, stetility!

Thanks for the advise! I've just picked up a few large 72qt. tupperware containers and will be making a glove box, and a mono tub.  I have a good feeling that a proper inoculation method will be the solution.

yes toss the jars. start again. also you need to pressure cook grain jars. you can't steam them.

Well, just did a bit of research on steaming grains... and it seems that it doesn't work well.  Mixed results at best.  Shoulda read up on it.  I had a successful Albino Penis Envy harvest with BRF, but wanted to try some truffles so I switched to grain, (pictured above).  Right now I have some Amazonian Cubensis I inoculated on Rye Berries about 3 days ago.  Steamed them, and used a glove box.  If it goes south a second time, I guess I'll know that the steaming isn't doing the trick.  But, I went by the book as far as the rest of the prep goes.... We shall see.

i'm betting those grain jars fail too. it's recommended to PC grain jars for 90 minutes to 2 hours.

edit: i'm sorry about my last post. i did not mean to imply that you need to throw away the glass jars. you just need to dump out the grain and wash the jars.
