I remember seeing in Strobel's paper that the media they used was a broth with the polymer suspended.

Quote:

mpranil DLF an anionic aliphatic aqueous PUR dispersion with 4% N-methyl pyrrolidone (NMP) (Bayer MaterialScience). Fifty-nine fungal endophytes were grown on solid PUR medium (PUR-A) containing 19 mM NaH2PO4, 33.5 mM K2HPO4, 7.6 mM (NH4)2SO4, 2.5 mM Na citrate, 250 μM MgSO4, 19 μM thiamine, 0.05% Casamino Acids, 147 μM FeCl3·6H2O, 14 μM ZnCl2·4H2O, 12 μM CoCl2·6H2O, 12 μM Na2MoO4·2H2O, 10 μM CaCl2·2H2O, 11 μM CuCl2, 12 μM MnCl2, 12 μM H3BO3, and 1.8 mM HCl. To 1 liter of this mixture was added 10 ml Impranil DLF and 15 g of agar. The polymer was added after autoclaving the medium to prevent deformation.

He had a good trade history. At first I was pretty angry, now I just hope nothing has happened to him, since he hasn't even been on since like nov-dec.

Easier to work with non sterile materials anyway, maybe I will mix up some liquid culture mix when I get home and throw some Pestalotiopsis in.

What i'd suggest is to take another approach and immobilize Pestalotiopsis inside alginate-Ca2+ beads to facilitate the contact of the fungi with the substrate.

A brief idea for what you you could do is put the Pestalotiopsis into a liquid culture with the suspended polyurethane then mix it with your alginate solution.

After that you slowly let the mixture drip into a CaCl solution with a burette, a Pasteur pipette or something like that. You will then form alginate beads with Pestalotiopsis and the polyurethane beads encased.

That way it doesn't matter if the polyurethane precipitate once you sterilize, just make sure to resuspend them before mixing with your liquid culture.

We'd have to test and see if the alginate beads would be stables with the polyurethane inside.

Hello.

I am a surfboard builder and I am rooting for for Pestalotiopis Microspora or fungi to help with the biodegration of polyurethane.

I would love to organize the surfboard industry to recycle old surfboards before going to landfills.

Hi!

I discovered the Indiegogo too late but I am an environmental engineer working on community workshop (e.g Bamboo bike building). Can I still get some culture shipped?
I would like to see if we can use the Pestalotiopsis microspora to recycle both the excess material with sand during shaping of joints and old bikes that are broken (and as a bonus if the bamboo or wood of the bicycles can be used  as fixating substrate).

Which also brings me to another question, beside the growth of the Pestalotiopsis microspora, what are the side products of the digestion of polyurethane? This in order to look at how to close the loop.

Best,
Ben

Hello

I just saw now on a project in Indigogo in 2014 where it was possible for 20$ to receive Pestalotiopsis microspora.

I find it veryr interesting. But now the project is closed.

Does anyone now where could I receive Pestalotiopsis microspora ?

I am in a city in south america, very dirty, full of garbages and plastics. My projecti would be to devellop a culture of mushroom in order to eat the plastic.

Thank you everyone

I don't have it anymore but Leighton does and is sending it out.  See the post above yours.

As far as I know no one has actually used this fungus to eat any plastic.

The original paper had the fungus eating polyurethane dissolved in water, which is pretty much unlike any of the plastic debris that people get excited about eating.    So I am not really sure if it works, did anyone do any experiments?

Hi all, I'm very interested in learning to cultivate Pestalotiopsis microspora in order to explore it's potential to help break down polyurethane. It's been a few years since there's been any discussion here about experimenting with cultivating Pestalotiopsis microspora and I'm just wondering if anyone has learned anything new about the cultivation process? Does the mycelium grow on grain? Is there something I can use as a bulk substrate where I can add in ground up polyurethane along with colonized grain? Has anyone figured out an effective method of adding a liquid polyurethane to agar? Thanks for any help!

Also, has anyone ever tried using a saprophyte to help stop top soil loss from erosion like Paul did here. mycorestoration of abandoned road

It grows readily on agar, I imagine it will grow on anything with sufficient nutrients. I gave up on my experiments though have some dried sporelated plates still kicking around to toy with at some future time.

It does grow well on grain.  It doesn't really eat plastic though.

Maybe liquid polyurethane,  but that's really stretching the definition of plastic and isn't useful at all.

Hello folks I have quite a bit of funding for a project to create a more effective plastic eating mushroom strain from pestalotiopsis microspora but I have yet to come across a culture I can say for certain is legit. If anyone has a way to source this fungus please contact me.
