Inoculating plugs/dowels

Hey all!

  So I pretty much have only worked with active species, never worked with medicinal/edibles past culture work.

I'm just curious about how/with what you inoculate plugs/dowels.

Can you inoculate with rye grain masters?

Or can the dowels be inoculated directly via agar wedge or liquid culture?

Or would I need to inoculate sawdust with grain master first and use this to inoculate the dowels?

I'm very familiar with culture work and grain to grain transfers, I'm hoping the skillset translates well to the edible world.

Any of the above will work. I recommend putting a little energy with the plugs. Hydrate with malt or potato before sterilizing.

Right on! I'd prefer to inoculate with grain spawn as thats what I'm most familiar with.

Thanks for the tip about adding malt extract/PD

I assume they work fine with either jars or bags yea?

What's everyone's preference on inoculation then I'd like to hear if anyone has the time

I just put them in Ziploc bags.if you use grain you can skip the energy a small amount of grain will do.

Unopened ziplocks are sterile but how do you get gas exchange during colonization?

Also I'm not sure I follow what do you mean "skip the energy, a small amount of grain will do"?

Eat just celery and you will find out why that works, In short you would spend more energy digesting it than you would get back.
Wood and plant fiber is dense, rich and hard to digest. No energy, no digestion, or in this case very slow growth.

The mycelium will take a while to properly accumulate enough energy to grow well on just plant fiber.

Grain spawn is full of quick to use starch, making a potent kick start for energy.

Cut a small hole in ziplock n wack some micropore tape over it.
I usually soak dowels for a week or two throw in jar with table spoon hydrated grain, Pc 15psi 90min
In sab cut/tape ziplock. Tip jar contents in,throw in an agar wedge,shake move around contents at 30/50% colinized.

On the energy i also tried soaking dowel in orgainc fert and skipping grain and it worked quite well.
Think Ferather suggested it.

Sample 1 has a greater surface area, in its quest for both energy and nutrients. Sample 2 is mainly on a quest for energy.
What is happening in sample 2 is the sheer production of new cells and more enzymes, on complex plant fiber.

Sample 1 will compete with contamination, sample 2 will most likely destroy contamination.

Grain is energy . Wood is basic substrate. That's why you supplement sawdust better growth more yield.Starting with agar or LC it helps to add some energy instead of straight wood dowels that have been kiln dried.As far as gas exchange never had a problem growing in Ziploc and I've had them in there for months.

My method is to boil the dowels for about 60 min. Bag them in Ziploc then microwave them until they inflate with steam about 6 times in 2 days. Then inoculated. Not conventional but I've not had problems. Foolproof would be 90 min at 15 psi in jar or spawn bag.
