Liquid Culture lids

Hey guys, I've been busy making jar lids for grain. I made some lids with just the silicone over a hole, but added a filter to this after realising the part about a vaccum and loosing the spores! Heres a picture, feel free to give me any advice, whats your method of choice? Thanks Spiggle

I do the same thing but use a quart jar so i can shake it a bit without getting the filter wet
Never put spores into a LC, waist of spores

Yeah change up that filter, it's gonna get wet and open up your LC to contamination, better safe than sorry

The white plastic lids are what I use. Because they're a single piece it's super easy to drop an agar wedge in.
The filter and injection port are very opposite each other. I can lay my LC jar on its side and also not get the filter wet

Thanks for your replies, So I have a quart jar from what your saying I may use that instead, so you can make a LC from agar wedge, i didnt yet know that! I guess the agar will be fast to colonize, i have a couple of agar dishes in the kitchen, is starting from spores not worth it?

Go spores>agar>LC. Spores are dirty, you can't look at a lc and tell it's clean. With agar you force the culture to grow in 2d so you can see and transfer away from possible contaminations, once you have a clean culture on agar inoculate your lc.

came here looking for this exact info... just knocked up my first plates with a GT syringe from a vendor, I have prints of GT, b+ (apparently this is a shitty strain?) and albino... kinda wondering how I should really start all this... I was just going to fuck with plates and transfers and not trip on fruiting until I have something worth it... not really sure honestly, kinda tempting to just MS some jars, but I want to do this shit right... is this the right way to go?

I now see my house filling up with plates... agar is cheap as fuck and so are the plates, barely made a dent in that syringe, I read conflicting things about isolation and im kinda at a loss in terms of finding that diamond in the rough without eating 30lbs of mushrooms... should I start bu]y just going for and selecting the fastest myc growth on my plates?

Out of any variety you can find potent genetics, you may luck up like I did once or it may take hard work. The quickest way to reliable potentcy would be a couple ways: put some spores to agar transfer them just enough to expand/know it's clean and grow out a few ms tubs with those plates. take several clones, grow out and test each clone. Or leave agar plates to pin, transfer pins to new plates grow out and test.

thanks for the tips! I definetly want to start isolating, but I obvioulsy also want fruits... I'm planning on starting with the bottle tek after a transfer or two from the syringe, I would suspect this works in my favor as i can dedicate a bottle(s) to isolating something nice... does that sound right?

If you want a really solid LC lid build...I recommend using my design:

it's really just like yours except there is a crimped brass plumbing insert with poly stuffed into it below the tyvek filter...the brass plumbing insert makes sure the filter will never get wet...i've used these lids time and and time again successfully for LC's from Agar...if you want an in depth simple LC manual...here's what I can offer you:MY Liquid Culture manual with LC Lid Tek

I use unmodded plastic lids for LC. So far so good. I fill my containers to half capacity. You gotta figure they have enough air in there and in the water, and have a tiny amount of passive air exchange. Haven't had any die yet.

Hell no lids perfect. I recently went back to baby bottles with micro pore double taped on the top and hole drilled on the side near bottom for a ship. Easy bottom access and i can cook 35 of them at a time with just a 16qt pc. Its the best way to test clones with agar as well

newb here, can you post a little about your tek and how you test your clones? I have some local cyans that im cloning and theres more to get, I want a really solid tek for dealing with them, thanks!

While all of that is true. Its not a tek. Its basically the no tilt lid tek but with baby bottles. I have a new born so i have a few 100 of them laying around. Your stilling going to clean up your cultures with agar, then just take a wedge and top it in like regular lc. I don't write teks because my English sucks
