Am I doing invitro wrong?

They are on 2inches of perlite inside a monotub setting with the bottom holes taped up and the top holes filled with polyfill. These have been incubating since 19th December 2016 and so far only 1 jar has grown ONE pin.

They get 12hours of CFL light and they are at a temp of 22C

They appear to be fine, no contamination on them, I just don't know why they are taking so long to fruit :confused:

Hey Rot,
When you say "incubating", do you mean to say they've been in the chamber as is since Dec. 19?  If so, there is an issue.  If you mean they've been colonizing since then and they've only been in the chamber for a couple days, just be patient...

Not sure why you'd have the bottom holes taped.

By incubating I mean they are just kept at a certain temperature. Right now we have a serious cold spell where temps are constantly in the 5-8C, for a Mediterranean climate that is seriously cold but yes they have been opened and in that chamber since December 19th.

What issue you think it is?

I think I know what my issue is. I stupidly forgot that I had plugged in my fan heater on the same timer that turns on/off the light so I had serious temp fluctuations. During the day it kept the room at a constant 22C while during the night it's plummeting to 5C :facepalm3:

Taping holes lowers RH.air has to flow through perlite and out the chamber to pick up humidity

I don't have a spare tote to make an SGFC, is there any other way I could get them to fruit ??

Wouldn't taping holes keep RH in since there is no where for it to go? How does it loose so much RH with taped holes?

Try putting a quart plastic storage bag on them and poking a couple holes in it.  Give it a spritz with your sprayer and put over jar.

I went out and bought one and just gonna transform it into an SGFC

I don't think the plastic tote over the jars will work since air needs to pass through the perlite

I've used shower hair nets in the past without any chamber at all and had great success. For fresh air you just pump the bag up and down every now and then to expel and suck in air. Easy.

I've tried with freezer bags before tied with a rubber band to the lip of the jar and then poked two holes on the top but I wasn't very successful and everything contaminated

I'm just gonna be a good little boy and follow SGFC advice

Thanks to Inocule's simple oat prep I'l be doing a shit ton of invitro so I'll benefit :super:

Yup 1/4'' holes, spaced 2 inches apart, all 4 sides, lid and bottom as well. Its hovering about 1.5inches above the shelf and has like 2.5inches of perlite.

Yeah I guess I could add more perlite but the shops were closed when I constructed this and only had a bag

I usually do monos, I'm good at them but I never spawned directly from jars and I wanna see what happens
