Mycelium might be out of breath after the 4th generation. It is suggested that you don't over do it. On the other hand, you will at worse loose only a jar by doing a grain to grain that doesn't turn well.
4 generations is my answer.

in paul stamets book MMC you can make one grain jar into 1000 jars just by doing grain to grain transfers. so my guess it much as you want.

its just zerohero. I should be a grain transfer nut but I believe you need the best flow hood or glovebox money can buy. I wouldn't take my chance with anything else(its not worth taking that chance IMO). growing mushrooms is very easy, its sterile is the ahrd part.

However, Stamets suggests not going beyond the 3rd generation on G2G transfers.

From TMC, page 51:
After transferring mycellium from agar to grain, further transfers can be conducted from these grain cultures to even more grain filled jars. A schedule of successful transfers from the first inoculated grain jar, designated G1, through two more "generations" of transfers (G-2, G-3 respectively) will result in an exponential expansion of mycelial mass. If for instance, 10 jars were inoculated from an agar grown culture (G-1), they could further inoculate 100 jars (G-2) which in turn could go into 1000 jars (G-3). As one can see, it is of critical imprtance that the first set of spawn masters be absolutely pure for it may ultimately inoculate as many as 1000 jars!
Heh, I had to read the paragraph a few times before I could understand it too.
OK, you got a colonized jar (spawn master). Use that jar to inoculate - say 10 more jars. That's G2.(If agar was used to inoculate the master, then this would technically be G1.)
The Gn jars can be used to inoculate how ever many more jars, a 1000 is possible, I guess.
I am not looking at a Generation to be a number of jars inoculated, but rather each time a Gn jar is used to inoculate another set of jars - that counts gfor a generation.
Sooo, I guess as I read it, you can have upwards of 1000 inoculated jars from a single agar culture, but not to exceed the 3rd generation of transfers.
You confused? Good me too.

one old man has 10 children. each have ten daughters (he has 100 grandchildren) and each of them has another ten children (he has 1000 great-grandchildren). he has spawned three new generations of his bloodline. after he dies (of natural causes) there are three generations of his family, numbering 1000 individuals.

I see your still a mod here. I like the explaination? thanks!
daywalker - I am getting it now. I am wondering why they aren't any good after the 3rd generation though. does it say anything about that.

well said, shirley! zerohero, I find nothing that specifically states why going beyond the 3rd gen. is not recommended. hopefuly someone will chime in with a reasonable explanation. edit: after re-reading shirley's analogy, I can see how going beyond gen 3 could cause some of the benefical traits of the original isolate to diminish.

ahh I see it too.
but I don't know if that would matter. the only thing that makes shirley explaination wrong(incorrect, different. use which one you want) is that mushrooms are asexual(sp?). therefore the grandchildren would be the exact same as the grandfather. so why would it degrade? from the change from jar to jar? or the time it takes to make 3 generations?
hmmm.... interesting indeed.
