The Maillard reaction is what you should worry about. 'Caramelization' isn't the right word for it. I PC agar for 15-30 minutes. You don't need to PC liquid media for longer times.

Well ... the culture slant agar seems to have settled instead of gelling.? What a pain. don't wait an hour after tubes are loaded to turn on PC...

IME, agar powder dissolves better in cold water. if water is too hot, and u dump the powder in, it seems to solidify and makes it hard to mix. better to start with cool and warm up as u stir.
it doesn't matter how long u wait either, even if it solidifies before the PC, its going to dissolve again during the cycle.
if I ever have plates or slants that don't solidify, its almost ALWAYS because I didn't add agar or I measured it wrong. even if I swear up and down that I did....so you know...don't discount that either

I've heard someone say (Pasty I think...) that PCing agar too long can cause nutrients to be cooked off....do you guys have experience with this?

sugars and amino acids get 'damaged' by the Maillard reaction. + Maillard reaction products inhibit the growth.

I will PC some grain tomorrow. I put in a some MEA to check if the effects are significantly (2 hours 15 PSI). It takes some time but I keep you posted.

Well that whole thing was off to a rocky start anyways! Soaked toothpicks picks seemed to make some brown crud, replaced them with soaked stir sticks..(nobody had tongue depresses. i dunno the pH of these wood things?) Cleaning out slants wasn't the most fun.
Sooo next time:
25g+ mda 500ml cold water mix well NO 1hr DELAYS because gross fauxpaw 20 min at 15psi
now the current grey area.... How should the slants be in the pc? Should the rack be partially submerged? Could a coffee can or jar be substituted for a rack? Also tighten screwlids tight for pc yah? jars with water, above water, also in water?

These glass tubes seem to be fine with tight lids. I just follow RR's DVD. They haven't cracked yet. Meanwhile Im totally having a Mandela effect moment here.. Been making agar for years and never remember anyone using a saucepan? I just put my thumb over the bottle and shook the jeepers out of it..

If you use liquid media there is no problem with tightened lids. I always PC my agar bottles completely closed.

Do you partially submerge your rack or jar? I looked at your tek in your sig. Nice write up. I made some plastic 20x75mm a few weeks ago using similar actions. These new ones are glass 20x150mm.
