Welcome to the thread!
There are 13 contenders, all have been isolated and are all originating from a multi spore B positive syringe.
As of recent agar wedges from each have been transferred to their own rye jars.
As yet to come the colonized grains will be mixed 1:1 with 50/50 GVC, placed in individual trays and fruited in a common environment.
Points will then be awarded for overall speed and yield weight.
Who do you think will come out on top?! Cast your Votes! Winner gets bragging rights and mad props for being a super OG swagmaster.
Pics of the rye jars to come in ~week.
Yea the names are stupid, but they had meaning at one time.

Well its been almost a week and the preliminary results are in! Type (R) is clearly in the lead. Types (WR, R*, R*, and U*) are one tier lower, with Type (SR*) not far behind. The others are quite slow, and unimpressive, but hey they might make massive fruits so ill still include them in the yield comparison.

Ill give'em a good equal shake and leave it at that for about another week.

No one guess any of those, but i didnt give any background, so kind of my fault. Colonization speed doesn't necessarily correlate to yield though, or does it?

I hope your wrong too! I dont think its bacteria, just saturated rye starch. Iv been told my grains are too saturated on a few occasions, and i think what your seeing is one of the byproducts.

I don't see anything wrong, just over saturation. Was it the last jar to be filled by chance? The bottom of the pot is usually wetter ime. Can't wait to see what comes of the fruits!

Were you using proper isolates for these? or just sectors from an MS plate?

Honestly suspect jar looks bacterial to me as well.

There are 13 jars total I only photographed the 6 best. Some of the others are just as wet but without the pancaked grains.

Your right it was one of the last jars filled.
Iv experienced the same thing before with most of the jars in my last project, which was moderately successful.
Seems to happen when grains get pancaked to the jar wall and spill their guts. The starches soak up water and become gelatinous blobs. Kind of looks like water from boiled potatoes.
The jar in question is clearly wetter then the other. I suppose time will tell, but Id be surprised is it was something more sinister.

The stuff on the bottom has been colonized, so i think its safe to say its not bacteria.

Good stuff, the way I prep my grains it happens to me sometimes with the last couple jars. It's never been an issue  I clean the grain, then pour boiling water over it and let it soak for 6 - 10 hours (stirring at least twice). Easy and works well.

weird, for me any standing moisture in a jar turns the uncolonized grain it touches bacterial before it can ever get a chance to colonize. So in my experience wet grain=bacteria
I always shake my jars a few times after removing from PC to distribute moisture back into the drier grains.
Have you grown out any of these strains before?
Hopefully you get a clear winner!

I would have thought that sterilization would kill off everything?
I'm guessing it originated from work in the SAB.
Upon closer inspection it looks to me like green mold. What do you think?

Depends on your technique. Some bacterial spores are very resistant. Some might even survive pressure cooking. Fractional sterilization is the way to go. Works even without the PC.

I would say it was introduced in the SAB. Bacteria can survive PC but as far as I know molds cannot.

1 lost jar is nothing to sweat over, and I agree with tump, black mold. It most likely came from SAB work, even if you were perfect, black mold is common in closets, bathrooms, and kitchens. But mainly crawl spaces that are lacking proper drainage.
I bet you did SAB work in a closet that has a wall adjacent bathroom or the kitchen?
I worked for a restoration company for a while, we specialized in killing black, yellow, and white molds. Shit was gross. The bad cases of black mold were always caused by a unnoticed leak or lack of drainage and dehumidifiers in crawl spaces. Insulation in crawl spaces would be covered in it, we would tear all that out and kill what was growing on the wood by spraying and rubbing Shockwave into the wood with rags (hopefully killing spores in the process). Then the team of painters would come in and do a sealant, then we would dig drainage ditches and replace sub pumps. I learned a lot about mold through that job. Gross as hell though, I do not recommend.

There is a vent right above where I used the SAB. Might have been a pressure difference push out some spores.

I like that observation, definitely could be a factor Your contam rate wasn't nearly as bad as mine is right now.. I have to keep the seal on my PC with clamps and it takes forever to build pressure if it even does so this round I've been crossing my fingers. Lost one tub of what I thought to be clean spawn to bacteria, lost another to trich (I think that was me on pasteurization), and a bunch of jars didn't even do anything because of the bacteria present I'm so close to that new PC....
Still have 3 tubs and a mini plus 6 clean looking jars that I'm going to spawn into more minis. Only one is fruiting right now though and the others should have pins already I'm pretty sure the one that is has a wee bit of bacteria too.
Awaiting an update! Something has to be brewing, Lets see the tubs!
