What's new, Shroomers?

I'm new to the hobby and the site but I've done a bunch of reading and I feel ready.


I started with some purple mystics syringes and some of Z strain.

Followed Shea25's PF tek and made 14 cakes. Half PM and half Z.



Next was Pastywhyte's Pasty Plates.  I made 24 and did half PM and half Z.
They turned out pretty damn stiff. I'll need to use less agar next time.


I bought some pre-made grain bags. Yawn.

And finally I did three LC jars following Cyber's LC tek.



Miscellaneous stuff



Everything except the plates are going just fine. Only 2 out of 24 show ANY growth.  Some have tiny black specs where the drop landed and some dont.

I figure I fucked up the sterilizing of the needle.  I read somewhere to alcohol wipe after flame to clean up the soot. Next day I read that I should so not have done that. At least theres no mold or bacteria.



I have no way to get any more agar for two weeks and I need to transfer that beautiful red plate. 

NOOB question: can I transfer to one of the plates that have no growth?

The needle may have been too hot when you inoculated. I figured that's what happened to me, killed some spores.

Add a prefilter to your FH blower.

Flame the needle push out 2-3 drops to cool the needle >>> incoulate the palte with one drop.

Do you use wide moth jars? If not you will have problems to get the cakes out of the jar.

That's what I was thinking. I cooled the needle with a drop and then wiped it.

I've got my pre-filter rolled up behind the blower and my jars are wide mouth. :thumbup:

How long ago did you inoculate the plates?

Some will disagree with the following advice and probably recommend you make 3 or 4 transfers before you cultivate anything with your culture.  But you can also do things a bit quicker. 

If you want to get started sooner you could take a transfer from the leading growth on that first plate and put into the LC.  The fact that you don't see contams on any plates shows that you have a pretty clean syringe.  When your new agar shows up you can test your LC before using.

Meanwhile you could also transfer to the plates that didn't show growth and start selecting for rhizo growth for the next grow. 

Buy plenty of agar so that you will be prepared to take clones from your best fruits.

I also had some problems with spore solution to agar.  Tried two times with 15 plates each time.  First time I just did a drop and when I saw no growth after two weeks, I thought the hot needle was the problem.  But the second time I squirted some solution out of the syringe before I inoculated the plates, and I also didn't use a drop, more like 10-20 drops all over the plates.  Again, three weeks later, there were no grow.  So I inoculated some mini-jars of grain and will transfer grains to agar when they're colonizing.


I never had any issues with spore prints to agar, only with syringe to agar.

So if you haven't seen mold or bacteria growth on those plates yet then I would bet they are clean, but there's still a chance that mushroom spores will germinate in the next week.  You could use them for making transfers too but you risk blending your transfer with newly germinated spores.

I think I understand what you're saying about mixing MS.  I'll wait another 5 days and if there is still nothing on the plates I'll transfer and I'll place the transfer away from the "drop site"
