How does this sound for a simple tek?
    #23965859 - 12/29/16 03:30 PM (7 years, 2 months ago)

soak rye berries over night, drain and refill water, simmer rye berries in pot until boil on low heat for 10 mins, then drain. fill 16 oz wide mouthed jar until 3/4 - 4/5 full, place tyvek over top of jar then metal cap with small holes punched for inoculation ports, then screw on lid. place jars in pressure cooker for correct time and temp/psi after completely cooled down then inoculate with syringe and place in incubator until colonization is complete.

Is there any improvements I can make?

I don't see how this is an improvement over existing grain prep teks.

Usually there is no need for an incubator. What is the temperature in your room?

why agar personally? I would liketo experiment with it one day, but for sake of cost I don't find it worth it right now.

if a spore syringe is made from 1 cap wouldn't the spores all be very similar already so that a test to see variation wouldn't yield different phenotypes as would be the purpose as i see of using agar plates? or is there a worthwhile reason I'm over looking?

Need to get rid of the inevitable bacteria/rival species that syringes carry and that agar makes visible

No need to drain that water.  If anything stir your grains up and pour off dirty water a few times. Add 1/4 tsp gypsum per cup of dry grain into final soak water Let sit for a day, short boil, drain, load. By incubation I hope you mean under 80F.

Consider upgrading to some rtv high temp gasket maker for self healing points and synthetic filter discs for gas exchange.  Shit is cheap af and significantly better.

You can do agar cheap too. Just need to read others methods. The sooner you get into that, the sooner you level up.

The spores dropped from a fruit have a very large range of genetics. The parent fruit doesn't really matter once it is stable enough. ( Unless your trying to cross varieties)

I used to just knock up grain with syringes all the time with great success. Then I started agar and now I feel dumb for not trying it earlier. You end up having many many more fruits per syringe then you would just knocking them up.

You also have the ability to see (most) contamination .

The list continues.

Utsf.
Look up cloning and pastyplates.
