First-time Agar question

I've never used Agar before, apart from a microbiology class taken in college, but I recently made up about 20 dishes and have a few questions. From what I've researched, Agar is used for isolation of a single sector, cloning, liquid cultures, etc. I used a MS syringe ordered from an online vendor to inoculate a couple dishes. This is what normal Mycelium growth looks like correct? (I used entirely too much MS solution on this plate I know, newb mistake)



My thinking is that inoculating a couple agar plates with the MS syringe, and then transferring the colonized agar into the grain, the grain will then colonize faster. Is this a typical use for Agar? I don't intend to isolate anything, or even do a transfer from one plate to the next (unless there would be a contaminate). I would just wait for the mycelium to grow enough on the agar to make sure nothing is contaminated, and then transfer to grain as long as everything remains healthy.


I guess I'm wondering if this would be a quicker way for grain colonization, rather than shooting the MS directly into the grain? Would this still be considered a 'multi-spore' grow since I'm not taking the time to isolate? Any help is greatly appreciated!

That growth looks clean but it's always a good idea to make at least 1 or 2 transfers after inoculating agar with spores to ensure your culture is actually clean. Going straight to grain with spores is probably faster but that's not the point. In my experience spore syringes are dirty more often than they are clean. We go to agar first to make sure we inoculate our grains with clean mycelium. Yes that's still MS.

Okay gotcha. Would you say now would be a good time to transfer to a new plate then? Or should I let it grow out more? I'm looking for the most rhizomorphic part of the growth to transfer correct?

I'd let it grow out a little bit more. Rhyzomorphic growth looks cool but doesn't really mean anything. I just go for the fastest moving section of mycelium that looks Farley organized. You don't want to take anything that looks too puffed up. That's why I'd let it grow a bit further from the center. If you're in a rush you could probably get away with taking some of the thin mycelium on the leading edge at around 3 or 7 o'clock.

Roger that. I'm not really in any rush so I'll let it grow out more. My original thinking was that placing pieces of colonized agar into grain would be a quicker way for the grain to colonize, rather than shooting the MS syringe directly into the grain. I'll do a couple plate transfers before inoculating the grain to stay on the safe side of things. Thanks again for the advice!

It's definitely the way to go but I wouldn't say it's quicker.

id say agar is only slower if you are starting from scratch.
once you get everything going it makes things way faster.

you can make LI or do g2g which take fractions of the time ms-grain does.

plus growing the cultures is almost as much fun as fruiting imo :stoned:
