Spore syringes and agar plates

Got a bunch of syringes in my stocking and made a dozen plates yesterday. Have a few questions for the masses.
When nocking up plates with a ms syringe how many plates will you do? 1 plate per syringe then make transfers or several plates per syringe then make transfers from the better plate?
Keep needles on or take them off for storage of partial syringes? I have always kept them on and flamed before and after.
Anything wrong with keeping clean ready plates in the sab until ready for transfers?
i keep my prints and syringes, both unused and partials, in a drawer. Should be good for a long while correct?

If you have 12 plates, I would do 6 & 6. only work with 2 different species/varities at a time. juggling multiple cultures can quickly become a pain in the ass.
keep needles on.
keeping them in the sab is fine.
keep them wherever. they last for years.

Yeah I had already decided to keep it at 2 varieties at a time since I will probably only have 2 tubs at a time. But I just did 1 plate each thinking I will save the other plates for when start transferring the myc. Any real benefit to makeing multiple plates from the ms syringe if you are going to make several transfers later from a single plate?

yeah... germinating spores isn't a gaurantee. its pretty common for some plates to have 0 growth... and it can take 2 weeks for spores to germinate. so assume you only make 1 plate of spores. and then you wait 2 weeks. and nothing grows. now you are pissed, 2 weeks in, and still haven't made any progress.
i would knock them all up, and buy some more plates in the mean time. what plates are you using?

Made up some pasty plates. I think i might as well knock up a few more just for the hell of it. I just didn't want to waste a whole lot of plates that way I have some ready to go for transfers or clones or whatever. I already have some cakes going and I also have some clean myc ready to go to grain. I just recently started agar and I can't wait to have a grow from it.

you're going to need 20-30 pasty plates eventually. the more the merrier.

thats fine he offered that advice thinking you are using petri dishes, not pasty plates. pasty plates are not wrapped, and the foil is fine if you removed it before inoculation, its only there for the PC cycle and nothing more.

One syringe per plate?? good luck with that.
Every time I germinate spores I hit up 5 plates at least, with a drop of solution from syringe. I mostly use prints though since syringes suck ass in my opinion but I still use 5 plates.
With the syringe I take the needle off and recap the syringe. I don't like leaving the needle on cause it can leak and cause problems.
I keep all my current plates in my sab unless I need the space. If I need the space I put the (parafilm wrapped) plates in a ziploc and set them outside the sab, do my work and put them back in if they are the next item on the list. If not I either leave them out or keep them in the sab until I transfer.
As far as storing syringes goes, I just put them in a cardboard box with other shit and stick them in a closet.
With pasty plates I can imagine you don't need parafilm or a ziploc, just put them wherever and wipe with iso when ready to use.
This is some easy shit op.
