I tried to clone a mushroom and it was not easy. I need help!

All right so I wanted to try to clone a mushroom. I prepared my SAB and flame sterilized my scalpel, but then I realized I don't have a good way of cutting the tissue apart. I was wearing gloves but didn't want to touch the mushroom to hold it down while I opened the stem to cut it out, but I did anyway because I didn't know how else to do it.


Also, I wasn't sure what to do it ON, so I used a ceramic plate from the kitchen which I cleaned with soap and water and then with rubbing alcohol.

I wanted to isolate and get only the tissue from the "inside" meaty part of the stem because it's most sterile there but it was not easy to take apart.

Should I use tweezers in one hand and a scalpel in another? Got any tips? Or teks specifically on how to take a tissue? Cloning an agar pin is easy enough, but a full grown mushroom, seems quite tricky. The worst part is that tissue I cut out never even grew on the agar for whatever reason it didn't revert to mycelium. :frown:

The Lower on the shroom the better, try a scraping motion rather than trying to cut out a wedge. If you scrape the tissue a bit of mycelium should stick to the scapel.

O shit, really? That's tight. Should I try to just split the stem open, reflame/sterilize and scrape the center/inside of it? Do I need to visually see that I scraped a significant amount or not really?

i hold the fruit like i would with a smartphone(the the fruit facing me)

and cut open the stipe at a thick part. i just scrap around inside the stipe
and pull out some tissue. its not like you need a big piece.

ive also been really sloppy and just cut it like a carrot and tossed it on agar.
needed to transfer a few times. diirty as fuck.

size of the shroom helps too. this guy was rather easy to clone..

this cluster was a bitch.

Tear the mushroom in half and use a very sharp exacto/scalpel to extract a tissue sample.  I clone where the stipe meets the cap.

Follow up question... how do you clone a cluster? Do you simply get some tissue from the stem/stiped of ONE of those mushrooms?  Or the base of it?  And if you do clone from that... it grows mostly clusters next time? And do clusters yield more or something?

I clone the bottom of the cluster. The point where the mushrooms stick together.

hah. ive only been doing the hobby for a few months. before that i was just
a guy trying to grow drugs. so i never cloned or anything until recently.

I cloned some clusters. You definitely will not get clusters always. The chance to get clusters is higher but it is not guaranteed IME.
I prefer to clone fat or tall dense fruits.

It looks like the environment/micro climate/conditions play a big role.

in HEPA flow, I split the stipe open in half.
Then fold the stipe over my index finger holding the ends with my middle finger and thumb... this really exposes the tissue you want.
Then I fillet a small (rice grain sized) piece of tissue and place into agar.

So Im holding the stem inside out with my one hand and holding the scalpel with the other hand.

I've not tried putting the sample down and cutting that way... always found it easier to hold in my hand.. more control over the fillet angle.
