First time using petri dishes, couple questions.

Hello! I purchased a premade pack of Petri dishes and am having moderate success. These guys are on day 8.

Feeling good about how these are going along I purchased some pp5's and MEA to attempt some pastyplates but had some questions. I tried making plates before and had messy results so I'm more than comfortable pouring after and try those later.

I was wondering if I could stack my containers and lids inside a spawn bag and seal them up how long would it take to ensure proper sterility?

Sterility got wrecked as soon as you opened those petri dishes. Unless you have a pimped out government lab to keep things sterile, with UV lights...etc then the best you can do is be as hygienic as possible and keep sanitation levels high

What are you working with? In front of a hood or SAB?

I keep my petris in a food container storage box. I open it infront of a hood and close it infront of the hood to keep things clean. I also wipe the inside of my box with iso, let it evaporate a little bit so it won't turn my marker markings into slush and keep it in a place with little to no air circulation to decrease the chances of air borne contaminants. I also keep the temps around 19.5 - 20C

You also should be wrapping your petris in parafilm or some other material to keep the contams out. I use masking tape cause I can't seem to find any parafilm that actually sticks

Those aren't open unless in a SAB where do you see signs of contamination? I use Ziploc bags, they work just as well as wrapping them up. They stay in a plastic container.

Anyways do you know how long I should keep my plates in the PC for, without agar on the plates?

I never said anything about them being contaminated. To me they look fine but I am not that experienced enough with plates, I started out a few months ago

I am not sure about plates in the PC, I had read somewhere once and to my recollection it was around 20minutes, however have a look through the search function, I'm pretty sure you'll find something useful

I am trying to work on isolations of B+, not going so well and it's a little disheartening but you gotta keep your chin up and always make back ups

Going to use this as a Q/A and log.
A good fourteen days later and I'm starting to see vectors to isolate on these first two pictures.

These two have some good growth.


I was wondering moreso when I would isolate to new petris, the sooner the better or should I let them grow out until they reach the edges or contaminate?

You can transfer now if you wanted to, but personally I'd grow it out some more, about 3/4 of the plate

I managed to make my 2nd transfers around that size as well, a pain in the ass :lol:

Listen to Kenetic and let them grow out a little bit

Happy New Years guys, made some traansferss a couple days ago and they're looking good.


I'm getting ready to jump into pastyplates to finish expanding on this culture but had some questions. The 1/4" hole needed for gas exchange is typically covered by several layers of micropore tape, could I substitute polyfill and have positive results?

Have a small issue, all ten of my plates are forming these blobs ontop of the agar. Wtf is this?

Was gonna say if all got the same contamination its from a fault you performed per dish so either like you said how you poured or inoculated or sterilized one of those three
Or sorry one more what environment did you pour in?

Also I don't use the bags that come with my petri mostly just wrap in parafilm and put on shelf with some space lol

Could you re-iterate the steps you've taken when pouring and inoculating the dishes?

It could help us pinpoint where you might have gone wrong
