Agar problems

Ok so what's the deal with sectoring. I made 4 dishes from a print from my last grow. After they grew out no sectoring. I figured there were so many strains that I would need to do it a few times. Took a small sample from the leading edge and spread it to 4 new dishes 4 diff cuts. Again no sectoring again I repeated. I'm on dish set 3 now and again no sectoring visible. I suppose I could post some pics. How many times will I have to do this. It says nothing about this in tmc

What recipe are you using for your plates?

You'll need to do a few more transfers before you see sectoring generally.

Post pics of your plates, preferably lighted from the back. Sectoring can be a bit tricky to spot. Also keep in mind that you are dealing with significant genetic variation. Depending on the spore count you started with, its possible that you still have a lot of variation at play.

Ya I'm having trouble getting good pics. But I'm examining them pretty hard and I have tmc as well have looked up countless pics and videos from here and rr videos on YouTube there's definitely no sectoring. I guess I'll just keep doing transfers. How big of a wedge do I cut? Nowhere have a read that it takes more than like3  or so transfers. Everyone made it seem like you'll have a mono by the third plate lol. I have blue tape on the dishes that wrapped into the back making pics pretty hard. But they look very clear holding them. Definitely no sectors. Should I cut as small a wedge as possible?

It can take many transfers from spores to see sectoring. I admittedly haven't read TMC, but there's so much we've learned since it was published. It might not help you at this stage of propagation, but this thread has great info on isolating a culture.

I cut a wedge about the size of a piece of rice or sometimes half that.

What's your agar recipe like, also?

Yes thank you for the link I read that once a while ago but now I will bookmark it. 30 grams pdy 750 ml water. I have a second type of agar as well but haven't used it yet. It says 25 grams mea 500 ml water

Do you have premix pdy or do you mix it yourself? When you make it yourself you can control the nutrient percentage. Less nutrients in the agar make the mycelium stretch out more looking for food, making it easier to identify sectors.

I have premix. Is there a way to water it down? Like adding water lol. Or will that not work

No, since it's a premixed you'll always have the same ratio of nutrients to agar. It's not the end of the world at all though, you'll still be able to get rhizomorphic growth with enough transfers.

Just get some grain jars going with your clean plates.  Spawn a mono tub.  Then when you get some good fruits take a clone.  Problem solved.
