I confess. Going back to the SAB wasn't as bad as I thought. My only real problem with it is the fact I can't see through it well. Might get a cheap piece of glass and make me a window in the front of that bitch. Haven't done anything with jars in there yet but honestly culture work was actually fun. Didn't have to go out to the cold garrage and got to sit down with some dabs in comfort. Hell I might keep doing transfers in there when I get a new filter.

Disclaimer: I have no used a flow Hood yet
I really enjoy SAB work. Put on some jazz, smoke a bit (away from SAB that is) and just lose myself. I have a room with no windows or vents that I use as my lab and can easily be in there for 3 or 4 hours without even realizing it.

My gloves caught fire from flame sterilizing too fast after washing with ISO.
Guess what, this mode of fire is not easy to put out with clapping!
I had to actually tear off the gloves and throw them on the floor
Wont be doing that again (maybe)

I probably shouldn't have laughed at that as hard as I did. Glad you made it out alive.
I never had that happen, but have had the flame creep up my tools much closer to my hand than I would have liked and made me think of exactly this scenario. I always try to take a moment to do a mental checklist while I let the ISO evap from my gloves.

How do you sterilize blades and other metal tools
You PC everything prior and never flame sterilize? Or you just mean you flame outside the SAB?

People flame sterilize outside the SAB to avoid creating movement in the air of the SAB from the heat difference or whatever you call it, created from using open flame inside the SAB.
I dont know how significant the difference between outside/inside is, I doubt anyone really knows, but the majority of videos of sab work I've seen, does sterilize the metal tool OUTSIDE the SAB.

Yeah I mean that is mycology 101 but you *could* catch your gloves on fire with iso still (flaming stuff outside the box), that's kinda why I asked. I have never done that before but I could see how it's possible if you aren't careful. So then I started wondering if maybe he meant he uses no flame whatsoever
What are you using for a flame?

Yeah I'm curious about the "no flame" thing too. I have heard of people autoclaving several blades and loops and just changing them out but I'm not sure what the process would be; details and such. Just curious really....

I PC my handles and use sterile blades for each plate. I can use multiple new blades carefully on the same handle in one session. In the future I will be re-sterilizing my blades as well. For now I am lazy and a bit wasteful, but it's so cheap I don't care too much.
Let's stay on topic. I confess I have been majorly slacking in agar work for almost a month.

I confess I was also confused as hell by that, but perhaps for the same reasons as when you typed it.
Confession #2 I was doing the same until I stopped doing the same. Now I have a buttload of plates that need to be tossed and need to start all over again.
