agreed. I'd have dried them and then did a little one gram (or so) control dose to see how they worked. How long did they sit?

I confess, I'm ready to kill my landlord again. We're having a cold snap/wave come through, and my internal temps are dipping into the high 50s at night. It's fucking up everything I'm growing right now... and I can't deal with this again like last year.
So, yeah, I'm house hunting. I'm over landlords.

Good luck with your house hunting ShaperDreaming. Itβs awesome having your own place... except being responsible when anything needs upkeep! π
I confess, even though it feels wasteful - I like a fresh blade!

Especially when Iβm taking clone tissue samples!

I always use a fresh blade, everytime I get into the sab. When they are clean and sharp stuff doesn't stick to the blade like it does after it gets tarnished and carbon buildup on it. Blades are cheap and recyclable.

Today I set up a space in the basement, and used my old SAB for the first time in years... Confession: Felt like a shaky handed noob. Couldn't see what I was doing. Hit the top of the box a couple of times. Probably fucked up half my plates.

After a week, 7 out of 11 dishes were covered in satellites and bacterial splooge.
I did another round of transfers a few days ago. Paid much more attention to technique and moving slow and smooth. It's been 3 days and all 10 out of 10 plates look perfect.

I confess I think I'm gonna get a bottle of antibiotic agar powder to aid in cleaning up cultures, after years of being anti-antibiotic use for agar... Seems like no matter what I do I have bacteria riding along with 2 of my cultures

Did you try hot pour or sandwich techniques?
Get an nice thick layer of agar between you and the culture, the myc should be able to more through it without the bacteria.

I confess! I decided to slow down my growing until I finish my house purchase. I'm tired of not having control over my conditions inside my building, and now I'm already dipping into the high 50s at night here and it's fucking up my cultures I'm going to still grow, but damn! I'm just super annoyed at all this, and I pay way too much money for my apartment to be this fucking complicated.

APE > PE
But, I found APE to be a huge pain in the ass to get looking good on agar, so be prepared for funky myc and many transfers

Same Idea but kinda in the middle of a hot pour/ sandwich; Transferring a wedge to a dish that hasnt fully hardened yet and bury it in the solidifying agar, discovered this one on accident but now I use it regularly for bacterial cleanup.

Oh! I try to do that for my clones because I've always found I get the best results when my clone starts "in" the agar. Good to know I should for transfers too Thanks.

Only if youβre concerned about bacteria or other hidden contams, I try to place the transfer upside down and push it down to the bottom so the myc is as deep as possible

That would work but Iβd just Just pour agar over the dish the culture is already in, no need to use another one.

Yeah but if you cut into the bottom layer of agar taking the next set of transfers you'd be potentially carrying that contam to the next dish unintentionally
I have put transfers in blank dishes then poured over them before...worked well enough to get fruits...but still want that antibiotic agar mix from fp just to have as a tool to use

Thats why its important to pour it deep as possible, and don't do it on a fully colonized plate that way you can still go to the leading edge. Thats one of the advantages of transferring into a still liquid dish, acts like a regular cleaning transfer but with the benefit of trapping the bacteria in the agar.
Basically same idea as what you are doing just slightly different approach and as long as it works it works.
Using a brf puck like agar can be helpful for cleaning up bacteria too, it has a harder time spreading than the myc then you can transfer back to agar.
If you insist on going the antibiotic route to save money instead of buying from FP go to your local pet store and buy the gentamicin they sell for fishes, you add it into the almost cooled agar and swirl before pouring, cant be cooked but Iβm pretty sure thats what alan rockefeller uses for cloning from wild tissue samples.
