I confess I cannot science a swab. I have long wanted to apply scientific rigor to the spores of the APE. Alas, they evade me; I've tried several times, each time no results, or worse. Is there a works every time trick?

I confess that I struggle with swabs too! I’ve had very little success just swabbing the surface ... After a couple of tries I end up sinking the whole dam swab tip under the surface of the agar and leaving it there and putting the top on plate. Generally these end up germinating, but I have to tease out good myce from some contamination sometimes.
I have not tried this yet but you can do it and call it out Tek if it works very well:
PsiloPsyChIn & Spotters Super Frickin Awesome Failsafe Swab Inoculation Tek
1. Take an empty 10cc syringe and fill it 2/3 full with clean water - leave one 1/3 air space 2. Wrap syringe in foil and place in a pint jar pointing needle up, plunger down. 3. Wrap a clean shot glass in foil and place in a bowl. 4. PC 2 & 3 for a minimum of 20 minutes at 15 psi 5. Set up your still air box or flow hood 6. Place swab, foil wrapped syringe, foil wrapped shot glass in SAB 7. Unwrap shot glass, place swab side down in the shot glass 8. Unwrap syringe and point needle at close range at the swab tip 9. Slowly plunge the sterile water out onto swab and into shotglass 10. Suck spore solution up with syringe and inoculate plates or jars...

Makes me wanna bust out that ape swab. Going to try this soon
I confess that swab has been sitting around for ages, it's time for action.

I’ve had luck pulling fibers off with flamed tweezers and depositing them on the plate.

If mine dont start germinating in the next week I'm gonna do that. I rubbed the inoculating loop over the swab pretty well I thought but who knows if there were spores on the loop when I streaked the plates.

Just use supplemented sawdust. Most people like to make blocks; shoebox and monotubs work too though. This is what I'm doing right now:

The second tub picture is some morels I'm trying to fruit shoebox style.
BTW I'm ignoring best practices here, check out the woodlovers thread and the gourmet pic of the day thread to see the work of growers who aren't just messing around.
