0ne out of twelve mini master slants. All treated the same, PC'd 45 minutes in the same jar that remained unopened until I went to use them...

I have recently had some contamination problems and I realized it was dirty air was being sucked into my petris via small cracks in my parafilm.
The small cracks are not enough. You also need temperature fluxuations.... heating up will displace gas out of the container through small gaps. Temperature drops will cause the gas inside to decrease in volume... sucking air in from outside of the sterile container. If you are unlucky that will include contaminant spores / germs.

I figured this out via the location of contaminants along the periphery of perfectly clean / sterile plates. On close examination I could see small gaps in the parafilm. I also know I get temperature swings... the rest is some rudimentary knowledge of physics and flow dynamics.

I have assumed here that the vial you are showing had some minute means of gas exchange and that it was stored in some place less than sterile.

Ah, good call. :thumbup:

I posted a settle plate pic not too long ago. Lots of shit floating around my room.

I've been thinking. I PC'd them with lids loose for GE in a mason jar, removed from jar (forgot that step in previous post) when cool enough to handle, tightened caps and left at an angle to solidify in a SAB.

Then, once cooled and solidified, back into the jar and lid replaced on jar.

I guess during the cooling period something could have been sucked past the screw cap's seal...

I think next time, I could try to tilt the whole sealed jar of slants. Then, when completely cooled tighten the caps :shrug:

I confess: Cordyceps are taking over my myco area.  I'm moving edibles outdoors, and I tossed all my ready to use cube spawn in a big diversity tub. Should be a quick, interesting experiment.  Too many houseguests this summer to be active with the actives.

I confess I was a bit apprehensive about agar, but after doing it it’s easy as pie. Used my first ever plates yesterday for three clone tissues and three print swipes. Got a cloning kit on the way, and a inoculation loop. Going to get cereal about this stuff :lol:

I confess I’m pretty bummed about these KSSS shoeboxes. Maybe I didn’t use enough spawn/sub or didn’t prep with enough water but all 6 shoeboxes are just putting out nice pinsets of really tiny fruits. Even uncased APE mini blobs in shoeboxes weren’t this small for me..

I figure they all may put out some decent second flushers but I almost don’t even want to bother harvesting any of these, not to mention deal with drying them

Ksss tend to be really short, but that's abort central. Try to bottom water. That's a killer flush to just lose like that

Made agar last night ..late.. and waited for them to solidify... went back for transfers and to wrap up the extras and they were all still liquid... turns out I doubled up on the malt and didn't put any agar in my MEagar :shrug:

Tonight I'll try again.

Nice on the green houses mentioned above - would love to see how you guys end up setting up!
