I confess I'm tired and cranky and hungry and I can't find my RTV which I need NOW because my agar is done steaming off and I needed to use 4 of the tall half pints which I haven't made lids for yet. Uggggh. Taking a break for a minute.

buy some plastic lids and leave them just slightly loose.... for next time.
I've had a pretty good success rate with unmodified plastic lids. Of course if you have temperature swings or drafts and spores floating around your jars you have a greater risk of contamination with this type of set up but like I said... I've had good luck with them.
to be honest I use my unmodified lids in a pinch. I use modified (with synthetic filter disks and injection port) plastic lids most of the time.

I found it but it was too late to PC my jars so I refrigerated them. The people upstairs get up early so I try to be nice. Anyone know where I can get more of the tips that come with the RTV. My tip always gets clogged and I have to pull the silicon out with a needle.

um unscrew that tip and push it out the wide end from the small tip. I use kabob skewers to push that solid piece out then screw the tip back on.

I've been using a syringe needle. Works ok. I think I'm just going to buy a new tube cause attempting to plug the tip with a needle cause I can't find the cap isn't working so well. Plus it's punctured on the side. I'm ok with it. It did me well and it's not that expensive to replace.
Just got my hole saw kit in the mail. Now I don't have to drill extra holes to make up for only being able to make 1" holes. Life is good. I got 18 jars of agar ready for transfers tonight and another 12 in the fridge to quickly transfer after that so I can get some clean plates to start over with. I have so much work to do to fix all this mess. I also still have all these damn dirty jars to be rid of. That's tomorrows fun project.
Decided to use the amazon and escondido spore syringe since I know they are both clean and from good genes to knock up the 9 jars I made a few days ago so they aren't just sitting.
I have some temperature issues. Sometimes the temperature dips around 68 in my house. Not sure if the spores will germinate in temps that low. Going to have to keep the space heater cranked. Clearly the people upstairs are trying to save money because they haven't had the heat on.

I'm not sure if it's fine for me though. I like it hot. Got used to keeping everything warm for the mushies and my bones and joints like the heat.

Speaking of bones and joints ... were you able to get an effective dose of Kratom down or is it just too nasty not to use capsules?

I'll try it again soon. I'll see if I can get it down in some strawberry banana smoothie. I just thinking putting it in coffee was a terrible idea. Just because it works for my Stamets7 doesn't mean it can cover the taste of kratom.
Also I'm really trying to figure out what the difference is between using soy and animal peptones in my culture medium is. How are they different?

As far as I know it's because peptone is a protein and any kind of animal protein agar is undesirable as it can sustain the growth of generally unwanted or dangerous pathogens.

Uhhhh... That sounds bad. So basically I'm just throwing a doormat out for all the bacterial baddies. Fantastic! Cause I can has smartness. I was thinking it might make a more inviting environment for bacteria but somehow the idea of attracting dangerous pathogens to roost wasn't in the forefront of my mind. I'll use up this batch of plates and switch over so I don't accidentally give myself an infection. Perhaps putting on the old face mask till then.

I honestly don't know for sure about animal peptone but for sure any kind of blood agar or meat extract agar can for sure support the growth of dangerous human pathogens

Hmmm... This is quite the conundrum since I'm starting to see that soy peptone is fucking expensive and I relly like my current agar formula. I get really nice growth.
