It's probably not necessary at all, but I wouldn't put any in the first jar if you intend on actually soaking the fruit for an extended period of time. You cut from the inside and conks are hard as wood so they're usually pretty good to just cut straight from the center. I've done it a few times with no bacteria issues or other contams

Throw them in the freezer overnight, the tissue will probably survive as they also survive in nature.
I did this with a maggot infested Hericium jar, worked perfectly.
You could also go back into the woods and do the toothpick method (stick it in at the edge, go back after a week or so, take it out, put in fresh ziploc baggie, put the colonized section to agar, clean up from there).

would that toothpick method work with storebought mushies? I was thinking of getting some shiitake cultures going since I'm bored but I'd also like to try this

According to RR, storebought shiitake's never really work well because of senescence already being present or close.

What about doing a spore scrape on agar, would that reset the genetics?

Good fruiting cultures of Shiitake are very hard to find starting from spores, that's why everyone just takes the cultures from the suppliers. There are some cheaper suppliers out there, if you want I could link you one in PM?

Fucked off another culture(time constraints). Moveing backwards. Good thing it's for fun and set backs are part of the game.

The post about shabby or chic was excellent.

because it would have been soaking in its own filth all night
a common practice for cleaning tissue samples is to dunk them in sterile water. ive done as many as 3 baths before to clean up a wild reishi fruit

Indeed, you basically do the same as with a spore print to spore syringe, you dilute the contaminants, only with spores, you can't just throw away the water so you add more.
