I find that cubes will fruit at lower temps than most of us think. I had cakes fruiting at around 55-60f for a while. They just fruited slower and the mushrooms grew a bit slower.

I confess i grew a monster shroom and wanted to clone it but didnt have any readily available agar. So I tossed that badboy into the freezer...never freezing fresh shrooms again. Once I took it out and let it thaw it turned into a slimey mess. i threw it into the dehydro and my fat 35+gram wet shroom turned into a pathetic nasty looking .7 gram stick i wouldnt feed to my worst enemy.

next time steam sterilize (or pc) a peice of cardboard and put the tissue sample on it. that will keep the culture occupied until you can get it to agar

You could also have cloned directly to Liquid culture, a plate of BRF cake substrate, Millet, or whatever the mushroom eats. It is not as 2D but if your mushroom is starting out clean you are likely to get a pure culture on the first attempt. My first grow ever I cloned to LC and it worked very well. For germinating spores I prefer substrate plates then grab a piece of mcelium from that to put on agar.

I confess that cloning directly to LC sounds very sketchy to me.
It may have worked for you that time but have you had repeat success with that?
It is very hard to get very clean samples of mushroom tissue (not impossible just more than a bit tricky). So I recommend putting that clone tissue on agar (or cardboard) then transferring the culture multiple times until you are certain you have a clean culture ... then LC

I confess my first clone I ever took was clean as could be. But I have lost a few clones since then by taking only one tissue sample thinking it would just be clean.

I confess, the broken rib finally healed and I hit the Thai pads and sparred tonight. Rib feels good. My lgt is sitting idly waiting for me. My poor lady. She has some friends from tyc waiting with her.

I've been lucky a few times too.
I'm no expert but here are some observations for those who are listening...
1) split the stipe open with gloved hands - do not cut it open. Even a clean scalpel can push contaminants from the outside of the mushroom into the core tissue you want to sample.
2) smaller is better.... I have learned from many failures that tissue samples larger than say a pea tend to rot. I go for samples that are about the size of a capital letter O on a typed page. If it is a small pin I'm cloning I might go larger if I'm using the whole pin or nearly the whole pin.
3) like comeback kid said... do about 3-4 tissue clones. If you are like me you will have 1-2 that look strong and as many as 3 that are weak or fail. If you do just one tissue sample, you are rolling the dice with that culture you want to preserve.

I confess that I let both of my LGT and Ghostie attempts fail. They were a bit dirty from the start, I got both into a second transfer but then I was busy with visitors and the second transfers were messed over by a bit of contaminant that came along.

I confess that I munched down a QP without doing any other projects, then moved to 2 cases of truffles.
At least my swab from theChief seems extremely viable. Lots of agar work this month

I confess due to not having the financial means I fruited two quart jars 1PE and one B+ without casing because they are about 10 days past consolidation. I have to move in 9 days so it cant wait anymore.

Just did my third transfers. These were my favorites. The peach agar is my leftover experimental agar. I made sure to transfer the rhizomorphic mycelium spots. The green is PDA. I have been having great success not contaminating my jars which is exciting. It must by m y obscessive cleaning. I did see some small contamination in a few but no mold. Just a few tiny bacterial dots. They kind of look like tiny dots of mucus. I made enough jars I didn't have to worry about opening those ones. All of my new agar is MYA. I just got a jar of peptone so I can make MYPA next.
The first culture is a pan. The next two are purple mystics.




I confess I made this nightmare by trying to clone an outdoor cube. I only took a photo so other people could see what pin mold looks like since it can look a lot like mycelium before the black dots appear. I'm pretty damn sure I got the pin mold because one of the bins I dumped outback was contaminated with it. This jar was opened outside then tossed in a trash bin. Looks like there is some other stuff going on in there too I can't identify. I think I will skip messing around with my outdoor cubes for now so I don't contaminate all my hard work. I can just as easily get cleaner cubes from my bin and practice cloning later. I can experiment more when I move and have more space.

Blackcat, I do NOT like how those PM look, keep an eye on them. To ME that looks like some mold with some rhizo mycelium trying to get away from it. Probably isn't but that would mean that those cultures are not that great. Transfer that strong rhizo growth. None of my PM looked like that so far.
