Not all contams are created equal. Did you test the lc before using it? You can absolutely have something get into the lc without noticing it. Don't rule anything out. Also, if there was any bacteria in your spawn overmisting can be a death sentence.

I confess, Tryp is right on the money with the LC contam comment. Every now and then i get cocky with my LC abilities and decide to forgo the test.... It doesn't always work. Hence the 20 masters i had to toss the other day.

I use Munch's tek. 1.5% should work, I've done closer to 1% a lot of times. You'll just have to see what works for you.

I confess that one time I was working at a friends house and the only space to work was the kitchen. My hand torch ran out of fuel 1/4 of the way through my session and I used to stovetop to flame my needlea/scalpels for the rest of the session. 0 contains were had that day

In a SAB or just depending on the flame's sterile field? I remember a Bunsen flame was good enough when i was learning microbiology in uni. But guys here swear that it's recipe for disaster. So I never attempted.

I confess, I havent been using gloves for awhile now. Pretty much since they went MIA along with TP, and ISO
just scrub up my hands real good, up to the elbow with hot water and soap.

Use a sterile loop to move some to a plate. Contams hide better in a 3d environment.
Verum, we arent talking about the conditions necessarily, my approach has been very hands off. Transfers, inoculation, and spawning is all the work I do. I occasionally mist smaller spawn runs, but trying everything all at once runs you thin

If my hood ever broke down and I couldn't afford to fix it I'd probably quit until I could. I can't imagine ever dicking around in a SAB again. So much prep, wiping shit down, putting all the shit away again fuck that. Flick a switch let's go to work.

I confess I didn't use flame sterilization, just wiped needles with alcohol between inoculations. Here's hoping my jars don't go to hell, or trich.

Made a batch of pasty-plates. Cleaned out the old agar. Decided the micropore tape was getting kind of grungy, so I peeled off all the old tape. Wiped out the dishes, poured fresh agar, closed them up and put 'em in the pc.
Taking them out after sterilizing, I realize I never put on the new mp tape.
