I cleaned up this thread a lot because failure isn't interesting. Let it be known that I failed, a lot. I don't want to make this thread go on forever though, so I snipped it.

Germination! Unfortunately, I can't take pictures of pasty plates as easily as I can with petri dishes. I've also put Ps. azurescens colonised woodchips to agar. I will use it to inoculate grains.

So you're insisting on firstly isolating a good strain even though making a grain MS at first would be easier? I've heard that cyanescens doesn't have much genetic variation (maybe that's not true but I have no way to disprove that).

I have a spore print. I just use agar to make sure it's clean. The spores are clean, I've confirmed that. But my own sterile technique is lacking and thus I unfortunately kept contaminating the plate before it could grow out to a nice size to inoculate a grain jar with. Honestly, it's for practice and also because I want to make sure I have 100% clean spawn.

How much, by Azuresens, colonised wood chips do you have? I was just thinking that if your sterile technique is lacking, wouldn't it be easy to inoculate some pasteurised wood chips with some colonised ones?

Scale up as it goes, to ensure the wood chips are getting colonised faster than any competitors could get a hold of the spawn.

I've never grown anything other than cubes, but from what I've read so far, it seems like a doable way.

Never mind me though I am here to learn.

Will follow this with interest!

A few, my dad bought a clean culture and has been messing with it. He very gracefully donated some to me. :biggrin: But the azurescens spawn is for his personal garden, he likes growing pot and such but he is pretty terrible with mushies.

Anyway, it's fair to say my sterile technique is on point. I don't know the exact reason why I got contamination in all my petris, but since I switched to pasty plates I haven't seen a single contamination pop up. And growth has been really good. I have three pasty plates with germinated cyanescens mycelium growing outward and two with azurescens woodchips just taking off. :yesnod:

Ps. azurescens woodchips spread out to the size of a small coin and then a satellite colony of... something appeared so I did some transfers on azures and cyanescens. I took the transfers a bit ago and they've grown out, I did grab the leading edge. You requested more pics of the mycelium so here they are. :cool:

Sorry, but what? The azurescens is obviously fucked, but there is lots of clean myc on there around those woodchips. The contamination popped up only 24 hours ago and grew that much in those 24 hours, that makes it easy to spot a contam. Obviously, I transferred the clean myc. The cyanescens is 100% clean and is from spore, if you had read the main post you would've known that and about my previous failures with agar. I streaked the plate, it didn't just grow from the middle. Also, the pink is food colouring.

Seriously, I know how to tell clean myc from contamination and these are most definitely not fucked. They have lots of healthy mycelium available for transfer. I even transfered the azures before anything sporulated.

Okay then. :smile: It's clearly an advantage that mold spores take some time to germinate, isn't it? Also, pretty brave of you to put unsterile colonized woodchips on agar. If I didn't see it I would bet my ass it couldn't work haha.

I made a blenderless LI using two agar plates (Ps. cyanescens) to 400 mL of water and used it to inoculate 8 L of spawn consisting of 80% grains, 10% woodchips, 10% saw dust. Below is one of the colonies used to make LI.

Could I ask you why you haven't used more % of woodchips in your spawn? In my humble opinion, grains should just be used as energy source to help the myc grow faster on the wood. At least that's what I do.

I hypothesised that the more grain spawn used, the higher the yield will be.

liloldme in his grow found that the higher spawn rate with a better energy source (i.e. grains) yielded better than a low spawn rate with a worse energy source, such as saw dust spawn or LC.

The spawn's purpose is to propagate mycelium for the main substrate. The main substrate usually has little nutritional value compared to the spawn. In this case, the main substrate will be a mix of soil, wood chips and saw dust. This does not have enough energy to produce tons of mushrooms.

Yeah and with his high use of grain he better feed it more wood in those areas or it won't establish and give weaker flushes as it dies. It's my understanding he left plenty of mushrooms though to rot back into the mycelial mats. I'm worried about the areas he cased with peat coz of pH.

Cyanescens jars are almost done, they look good. I think I made a mistake with the saw dust, they don't seem to want to colonise it very well. In places they grab onto it, but it's pretty rare. Either way, shouldn't matter too much.

The azurescens jars also looking very nice.

Although I inoculated both jars with a blenderless LI I haven't seen the crazy growth I was expecting. Not sure what's up with that. I presume it's my hard agar, next time I will try soft recipe.

Any idea why they don't colonize the sawdust? I've heard of it not being a good substrate before, but I would like to know the reason. Nice jars btw. :smile:

Well, I found out it was spruce wood. (Norway spruce or Picea abies)

I think that might have something to do with it, but I also read multiple sources that it should work. I used the same saw dust with Reishi mushroom and they did fine on it.

Has been a long time since an update!

The cyanescens patch has been laid. :smile:

About 5 liters of spawn, into a 45 L monotub, kept feeding it wood over the weeks and eventually it was almost entirely full. I decided then it was time to bring them outside. Dug a big hole, put cardboard down in the bottom, put my last jar of spawn over the cardboard. Spread out all the wood from the tub into the hole. Covered with woodchips, few shovelfuls of dirt and mixed thoroughly.

The azurescens patch has been laid next to the cyanescens patch. About 4 L of grain spawn went into this baby. I soaked them in water and shook before inoculating the patch. Kinda like making a GLC but throwing the grains in there too. I think this will benefit colonisation. Thinly cased with mix of coir/dirt (the dirt is more like clay where I live :lol:). I want some plants/grasses in the patch, any idea how I could set that up? I don't think they're capable of growing there because the ground is pretty much solid woodchips.



I got Ps. serbica on agar but the growth is very slow. I'm thinking it might be bacterial but it doesn't look like it. Gonna make more transfers to get good growth, maybe I can get some fruits this fall.

Go for a walk, find some tall grass from last year, grab seed tops and spread...  I would think some grass would help with potency but that is only hypothetical

I don't think there will be any flowering grasses for a while... But good idea! I'll try to grab some when I see them. Do you think they'll grow on ground like that (with the amount of wood that's there)? Why do you say it would help with potency, I find that very unlikely.

Azurescens are described as the "most potent"  They grow amongst high tryptamine dune grasses.  Grains are grass fruits, also high in trypomines/tryptophan :shrug: I have been posting the shruggy guy a lot lately (edit) find an unmowed field with grass from last year rip the tops off or get some local hay from the farm store and mulch

The reason azures like dune grass is it is very high in lignin.

Throwing just any grass on there won't do didly to help potency.  Azures are just fine with fruiting on plain wood chips.  In the wild Azures fruit on the old decaying bits of European dune grass at the base, they do not fruit on the seed at the top of the plant.
