Whattsup everybody peeps.
So I inoculated a few petri dishes of agar 9 days ago. 2 of the plates have been showing some kind of growth for 4 days now these being of the PE variety. The growth does appear to be mycelium from what I can tell but is still very small and is growing very slowly??
After inoculating the petri dishes I immediately wrapped the plates around the edges with saram wrap. I am wondering if this was incorrect and if you only do this with plates which you are storing for later in the fridge. Could the fact that I wrapped the plates be slowing down colonization of the agar? Should I have perhaps just put a couple pieces of tape on either side of the plate to secure the lid instead of all out wrapping it?
Or could this be due to the fact I used pure honey in my agar recipe? I later read in another thread that perhaps using pure honey slows down colonization in agar due to antifungal properties in it, is this true? Should I going forward only use Raw sugar? Or does this in fact not matter.
Preferably looking for answers from a TC.

You inoculated with spores 9 days ago? It can take up to a week for spores to germinate so the growth doesn't sound too slow to me. The honey may slow it down, how much did you use? More nutrients in general will also slow down growth, if I recall correctly.

I here what you're saying about the 9 days not being too long to show signs of germination but these plates were showing signs at Day 5 and since then the growth has been very slow. I can make out the white spot of what to me looks like mycelium with a slight splatter stain around it likely from the drop of spore solution, also another spot popped up closer to the edge of the plate I may have accidently put two drops on that plate or it's bacterial time will tell.
This is my first attempt with agar so I guess I don't have much frame of reference...and like Donny from The Big Lebowski I'm a little out of my element at this point in time.
Just curious if the saram wrap around the edge makes any difference or not.
Honestly have no idea about pure honey slowing things down as it's not really in the teks I looked at in fact is included in many of them. Next time I'll go for raw sugar I guess.

It's really hard to take and share these photos, but these are 2 petris inoculated with spores on 14/12, one week ago. This is on standard MEA.

The mycelium appears translucent in growth and later turns white and thicker. As you can see, I use saran wrap.

Hey Tiamo,
You could put them on something dark for contrast as opposed to a white surface.

I'll see if I can get a good shot of what I have so far later today and I'll post it. I will be posting to my journal anyways. May just make a new batch of agar today using raw sugar in the recipe for my transfers.

Waaaay better! Looks like some good germination. Whenever I drop on agar I roll it around (streaking) to spread the spores around. Looking good there!

Ok so here are two plates that are showing signs of life.
The one I was discussing before is shown here labelled " PE 12/13 #1" like I said started to show signs of growth after 5 days.

And then here is another PE plate that just started showing signs today I believe.

So for Dish#1 would you say this is about normal for 4 days of growth or kinda slow? Also is it too early to say if it's mycelium?

Yeah PastyWhytes idea genius! Should definitely help spot the shithead colonies from the good stuff.
Hopefully I get lucky and don't need to do any transfers on one of these plates who knows.

I'd still recommend transferring it. Contams may not always be spotted. I believe the general idea is 3 transfers whenever you start from spore to isolate and get clean growth.

PastyWhyte advised me a while back that you can usually get a clean culture with 1 transfer so why waste the extra resources( agar, petri dishes) when you already have obtained a clean culture no? I can always grow it out large enough that I would be able to tell surely
