How I do no-tilt liquid culture jars

I basically use carbide burs bought at walmart to drill a hole on the glass of the mason jar towards the bottom so that it can be easy to draw liquid culture with a syringe...I hope this helps some. let me know what you guys think!

These are carbide burs. Diamond cuts glass pretty good too.

fill mason jar with water and place lid. Fill bucket or pot with water, just enough to come about half an inch above the glass area you will drill. (if you try drilling glass without water present it can cause powdered glass to get everywhere. so clean up is easier with water

Be very careful using anything electronic near water. I can not stress this enough! Put firm pressure and you should drill a hole within 5 minutes. Too much pressure cracks the glass. It may take a few shots to calibrate yourself to find out the optimum amount of pressure to place while drilling.

Fill the hole with RTV and let it set. Then prepare your lid with a hole and tyvek/SFD to gas exchange.

Final shot!

I recently made a jar with the hole drilled into a bottom angle/vertices of the glass so that I can tilt liquid into that corner and collect. It has made it so much easier for me to draw liquid cultures without having to make special tubes that come from the lid and reach into the bottom.

I did break several mason jars trying to figure the proper amount of pressure to apply and how fast I should drill the hole. I believe you can drill a hole in about 3-5 minutes without breaking or cracking the jar. the harder and faster you want to drill the higher the chances of breaking it. Just put on a cool song and by the end of the song you will probably have a nicely drilled hole. Btw, regular drill bits suck assss to drill through glass...either get diamond burs or a carbide bur like the ones I pictured above.

Dude sweet idea!
How do the jars hold up in the PC post drilling? Have you noticed any increase in cracks or anything like that? I imagine it would weaken the glass quite a bit, but sweet tek nonetheless man!

so far none have broken due to pressure cooking. although one day it may crack earlier than if I had not drilled a hole I think it's totally worth it for it's convenience. Thank you for the support

This could potentially be used in the following way:
Grow a culture on agar in the bottom of that jar. Inject sterile water into the jar and swish around to make a LC. Easily draw LC with a syringe through the bottom injection port.
This way you don't have to uncover the lid exposing the agar to possible airborne contams.

Most excellent idea, dude. I am actually going to walmart just to get these bits as soon as this posts... One question---have any experience leaving things in with the agar to assist mixing, such as a mag stir bar, a Blender Bottle Thingy, a screw or a marble or piece of glass ( as in wondering if that would poss effect growth on the agar?)

For liquid culture ive used broken glass cuz rusty nails cloud the water. But for agar i would put anything to stir. I just received magnetic stirrers but have not tried yet!
