Nice clump of spores clearly visible in each syringe. I was surprised that the PE one had so many considering they are apparently so hard to get prints off of.

You guys think it's best if I'm inoculating directly with the spore syringe to make an indentation with the tip of the needle first before leaving a drop in the middle?
Just use one drop or go for two or more in each petri dish?
Any advice would be appreciated essentially first time trying to germinate on agar with fresh syringe.( last time was using old syringes which were dead it appeared)

PE is hard to get prints, but not spores. They make tons of them, they just don't drop. Its a mutation that prevents the spores from ejecting from the basidium. However, you can collect the spores just fine with swabs or a centrifuge.
1 drop is all. shake it up nicely before you do the drop. an indentation is fine, but not required.
use less agar in your recipe than you normally would. if you don't have a recipe, use my 1-2-3 agar recipe in my sig, its perfect for spores as is.
drop 1-2 drops of solution on to the bottom of your sab before you put the needle over the agar and inoculate. after flame sterilizing your needle, you'll find it likes to spit out a bunch of hot steamy liquid for the first drop or so. you dont want that drop to be what goes on your plate.

Nice I pretty much do exactly what you are saying here to begin with. I will be using regular petri dishes though seeing as how I just got some sterilized ones the other day do I need to add or subtract from the recipe you figure if following your 1-2-3 method? Goal is to get at least 8 petri dishes out of the batch( 4 per variety) ?
